MKK6 expression analysis has been reported in various primary tumors, like esophagus, stomach, colon and prostate cancers but its significance in primary breast tumor patients remains unclear. We investigated the expression of MKK6 in cancer tissues through immunoblotting and immunofluorescence techniques in 25 resected cases of human breast carcinomas. Increased MKK6 protein expression was observed in 56% (14 ⁄ 25) of breast cancer cases. These results indicate that the upregulated expression of MKK6 in cancer tissues has significant role in tumor progression and the clinical prognosis of patients with primary breast carcinoma. …………………………………………………………………………………………………….... Introduction:-Breast cancer is one of the most common cancers with more than 1.3 million cases and 0.45 million deaths each year worldwide [1]. It is the number one cancer amongst females accounting for 5.59% of total cancers in females of Kashmir valley only [2]. Breast cancer development involves progression through chain of intermediate processes, which starts with ductal hyper-proliferation, followed by subsequent progression to carcinoma in situ, invasive carcinoma, and finally into metastatic disease [3]. Studies have shown that metastasis is a series of diverse events, generally known as a metastatic cascade. These steps include fleeing of malignant cells from the site of primary tumor, propagation and shifting to a discontinuous secondary site(s), and lastly, the survival and growth into clinically detectable metastases, a process termed metastatic colonization [4]. In order to significantly augment our understanding of the metastatic process and to identify the specific targets for cancer therapy we need to single out the proteins and signaling pathways necessary for regulation of metastasis. In various cancers many proteins show differential expression and regulation [5][6][7]. One such protein is mitogen-activated protein kinase 6 (which from now onwards will be referred to as MKK6). MKK6 belongs to the mitogen-activated protein (MAP) kinase super family and has a role in both apoptosis [8,9] as well as proliferation [10]. MKK6 has been implicated in regulating tumorigenesis. MKK6 has been shown to play contradictory roles in suppressing metastatic colonization in human ovarian carcinoma [11] on other hand it has been shown to be up-regulated in prostate cancers [12]. As of now, very little is known about the regulation of expression of MKK6 in the cell [13]. MKK6 activates downstream all the isoforms of p38 MAPK [14]. P38α has been implicated in regulating cancer progression, by regulating invasion, inflammation and angiogenesis which are key oncogenic steps in tumorogenesis [15][16][17].

Nowotwór jest schorzeniem, które powstaje w wyniku załamania się mechanizmów kontrolujących prawidłowy wzrost i różnicowanie komórki. Z procesem tworzenia przerzutów nowotworowych ściśle związane są zaburzenia polimeryzacji aktyny. Do najważniejszych czynników regulatorowych polimeryzacji aktyny należy wieloproteinowy kompleks białek WAVE, w skład którego wchodzi między innymi białko Abl interactor1 (ABI1). Uważa się, że zrozumienie roli białka ABI1 może w przyszłości pomóc w poznaniu mechanizmów koordynacji polimeryzacji aktyny i proliferacji komórkowej, które ulegają zaburzeniu w komórkach nowotworowych. Badania wykazują, że w przebiegu chorób nowotworowych ABI1 odgrywa podwójną rolę: wzrost ekspresji tego białka wpływa na rozwój i progresję inwazyjnego raka piersi, raka jajnika, raka jelita grubego, a także przewlekłej białaczki szpikowej i ostrej białaczki limfoblastycznej Ph+, z drugiej strony wpływa na supresję guza w nowotworach żołądka i prostaty. Analiza wyników badań próbek tkankowych pacjentów po gastrektomii z powodu raka wykazała silną zależność pomiędzy ekspresją białka ABI1 a stopniem zróżnicowania nowotworu, zaawansowaniem procesu nowotworowego oraz liczbą zajętych węzłów chłonnych. Należy podkreślić, że zwiększenie poziomu ekspresji ABI1 nie jest ograniczone tylko do procesu nowotworzenia, ale można je zaobserwować także w przypadku zmian zapalnych i innych bardzo wczesnych zmian rozrostowych, które niekoniecznie muszą transformować w kierunku raka, jak na przykład polipy hiperplastyczne. Dalsze badania ekspresji ABI1 na poziomie RNA i białka umożliwią ocenę jego wartości prognostycznej i wyjaśnią, czy może on być celem interwencji terapeutycznej. The literature reviewCancer is a disease that occurs as a result of the collapse of the mechanisms that control normal cell growth and differentiation. The process of metastasis is closely related to impaired actin polymerization. One of the most important factors that regulate actin polymerization is multiprotein WAVE complex, which includes, among others, Interactor Abl protein (ABI1). Understanding the role of the ABI1 should help to define mechanisms for coordinated actin polymerization and cellular proliferation, which are dysregulated in cancer cells. The dual role of AbI1 function is observed in human cancer. Increased ABI1 is linked to enhanced oncogenesis in invasive breast cancer, ovarian cancer, colorectal cancer, chronic myeloid leukemia and Ph positive acute lymphoblastic leukemia and to tumour suppression in gastric and prostate cancers. Analysis of the studies results of tissue samples of patients after gastrectomy for cancer showed a strong correlation between the expression of ABI1 protein and the degree of differentiation of the tumour, the advancement of the cancer and the number of positive lymph nodes. Proximity proteomics reveals role of Abelson interactor 1 in the regulation of TAK1/RIPK1 signaling

Dysregulation of the adaptor protein Abelson interactor 1 (ABI1) is linked to malignant transformation. To interrogate the role of ABI1 in cancer development, we mapped the ABI1 interactome using proximity‐dependent labeling (PDL) with biotin followed by mass spectrometry. Using a novel PDL data filtering strategy, considering both peptide spectral matches and peak areas of detected peptides, we identified 212 ABI1 proximal interactors. These included WAVE2 complex components such as CYFIP1, NCKAP1, or WASF1, confirming the known role of ABI1 in the regulation of actin‐polymerization‐dependent processes. We also identified proteins associated with the TAK1‐IKK pathway, including TAK1, TAB2, and RIPK1, denoting a newly identified function of ABI1 in TAK1‐NF‐κB inflammatory signaling. Functional assays using TNFα‐stimulated, ABI1‐overexpressing or ABI1‐deficient cells showed effects on the TAK1‐NF‐kB pathway‐dependent signaling to RIPK1, with ABI1‐knockout cells being less susceptible to TNFα‐induced, RIPK1‐mediated, TAK1‐dependent apoptosis. In sum, our PDL‐based strategy enabled mapping of the ABI1 proximal interactome, thus revealing a previously unknown role of this adaptor protein in TAK1/RIPK1‐based regulation of cell death and survival.
