BackgroundThe contemporary peak of diabetes seems to be related to obesity, sedentary lifestyle and diet. Diabetic retinopathy is the most leading cause of blindness in adulthood in industrialized countries. Our purpose was to evaluate the effect of a high-fat-diet (HFD) on the retina of diabetic rats.MethodsTwo groups of Wistar rats were injected with streptozotocin (STZ) two days after birth using 45 and 90 mg/kg, respectively. At 8 weeks the group on lower doses started to be fed on a HFD. Animals were sacrificed at 37 weeks of diabetes. A control group was made up of non-diabetic rats. Retinal flat mounts were examined using the trypsin digestion technique. Pericytes counts were compared between diabetic and control rats. Cross retinal sections were analyzed by histological techniques and immunohistochemistry and immunofluorescent technique. Primary antibodies against inflammatory and proangiogenic mediators such as RAGE, GFAP, 5-LO, VEGF and TNF-α were used for immunohistochemistry and Western Blot (WB) analyses.ResultsIn the two diabetic groups we observed GFAP-positive cells with a morphology and spatial organization similar to those seen in Müller cells. Both diabetic groups had a significantly lower number of pericytes than non-diabetic animals.Increased retinal immunoreactivity of GFAP, RAGE, TNF-α, VEGF and 5-LO was seen in diabetic animals fed on HFD compared to the other groups of animals. WB analysis revealed a higher expression of 5-LO, VEGF, TNF-α and RAGE in the retina of diabetic rats on HFD than in controls and diabetics fed on a normal diet. The percentage of RAGE-stained ganglion cells and ganglion cells was found to be significantly lower in animals on a HFD than in the other animals.ConclusionsDiabetic animals fed on a HFD showed an increased upregulation of inflammatory and proangiogenic markers. This animal model may be useful to study mechanisms of diabetic retinopathy and therapeutic targets.

The purpose of this study was to prove that the administration of Jati (Guazuma ulmifolia Lamk) leaves extract improves the lipid profile of dyslipidemic male Wistar rats. Subjects were 20 rats (Rattus norvegicus), male, Wistar strain, dyslipidemia (total cholesterol ≥ 200 mg dl-1), aged 2 months old, weighing 180-200 grams. The control group (10 rats) were given a placebo of 3 ml aquadest (P0) and the treatment group was given extracts of the Jati (Guazuma ulmifolia Lamk) leaves extract of 25 mg kg-1 BW (P1). Before and after treatment for 14 days, total cholesterol, triglyceride, LDL, and HDL levels were examined. The results showed that in the P0 group there were no changes in total cholesterol, triglyceride, LDL, and HDL levels (p>0.05), whereas the P1 group experienced a decrease in total cholesterol, triglyceride and LDL levels (p<0.05) and an increase in HDL levels (p<0.05). The results of this study indicated that the Jati leaves extract was effective to improve the lipid profile of dyslipidemic rats. It was necessary to compare the effectiveness of Jati leaves extract with synthetic dyslipidemia drugs used in the community such as statin.

This study aimed to examine the effect of catfish oil intervention enriched with omega 3 on lipid profiles and ox-LDL levels in the elderly. The research design used a randomized-controlled trial (RCT) in a single blind. Subjects were divided into three groups, the first group was given soybean oil (SO), second group was given catfish oil (CO), while the third group was given catfish oil enriched with omega 3 (CO+omega 3). The lipid profile and ox-LDL measurement was performed before and after the intervention. The results showed that CO+omega 3 group had significant reduction in total cholesterol. Triglyceride levels in CO and CO+omega 3 groups decreased after intervention, while triglyceride levels in SO goups increased, but no significant differences (p>0,05). The HDL levels increased in SO and CO groups, while CO+omega 3 decreased slightly, but did not different significantly (p> 0,05). LDL levels decreased in all groups, but the highest decrease was in the CO+omega 3 group. The ANOVA test showed that delta LDL levels in each treatment group did not different significantly. The ox-LDL level in each treatment decreased, but it was significant only in the treatment of catfish oil enriched with omega 3. Therefore, catfish oil enriched with omega 3 was chosen as the best intervention in improving lipid profiles and oxidative stress of the elderly. Contribution/ Originality: This study is one of very few studies which have investigated the effect of catfish oil intervention enriched with omega 3 on lipid profiles and ox-LDL levels in the elderly.
