Dynein light chain rp3 acts as a nuclear matrix-associated transcriptional modulator in a dynein-independent pathway

Abstract: Cytoplasmic dynein is a motor protein complex involved in microtubule-based cargo movement. Previous biochemical evidence suggests that dynein light chain subunits also exist outside the dynein complex. Here we show that the dynein light chain rp3 is present in both the cytoplasm and the nucleus. Nuclear rp3 binds to and assembles with the transcription factor SATB1 at nuclear matrix-associated structures. Dynein intermediate chain was also detected in the nucleus, but it was dispensable for the rp3-SATB1 inte… Show more

“…This analysis revealed matches with dynlt1/3 dynein light chains (DLCs) of D. melanogaster and C. reinhardtii strongly suggesting that MGG_01005 is the M. oryzae orthologue. Cytoplasmic dynein is a microtubule-based complex involved in a variety of cellular activities 23 25 . It comprises two heavy chains (DHCs) and two or three intermediate chains (DICs), light intermediate chains (DLICs) as well as DLCs 26 – 29 .…”

“…Interestingly, the distribution of L2 in the dynein light-chain knockdown cells strongly resembles the Golgi-localized L2 and pseudogenome described in this study. Intriguingly, the dynein light-chain rp3, which shares 55% homology with Tctex1, has been shown to associate with the nuclear matrix, where a subset localized to ND10 (53). Tctex1 has also been found in interphase nuclei (54).…”

“…DYNLT1 also interacts with Lfc, a guanine nucleotide exchange factor in a phosphorylation dependent manner to regulate neurogenesis [69], [70]. RP3, a dynein light chain subunit, localizes to the nucleus and is involved in transcriptional regulation [71]. Localization studies showed a sub-population of DYNLT1 in the nucleus [63].…”
