Dynamics of Choline-Containing Phospholipids in Traumatic Brain Injury and Associated Comorbidities

Abstract: The incidences of traumatic brain injuries (TBIs) are increasing globally because of expanding population and increased dependencies on motorized vehicles and machines. This has resulted in increased socio-economic burden on the healthcare system, as TBIs are often associated with mental and physical morbidities with lifelong dependencies, and have severely limited therapeutic options. There is an emerging need to identify the molecular mechanisms orchestrating these injuries to life-long neurodegenerative dis… Show more

“…The treatment of epilepsy has been a huge challenge for neurologists due to the high refractoriness of current therapy ( Javaid et al, 2021 , Javaid et al, 2021 ). The mainstay of therapy targets not only to protect the patient from seizure episodes but to improve the patient’s overall well-being by maintaining the psychosocial activities and evading the toxicities imposed by long-term drug use ( Goldenberg, 2010 ).…”

“…Epilepsy is one of the most common central nervous system (CNS) disorders, with an incidence of approximately 1% of the population. Post-traumatic epilepsy as a comorbidity of traumatic brain injuries is also becoming a universal challenge for brain health due to the increasing incidence of brain trauma [1]. About one third of patients eventually develop drug resistance to some anti-epileptic drugs (AEDs) and fail to respond to treatment with medication [2], although a variety of combination pharmacotherapy of multiple drugs was investigated [3].…”

“…Choline plays a prominent role in the synthesis of different membrane phospholipids (Javaid et al, 2021). Moreover, choline is essential for cholinergic neurons to synthesize the neurotransmitter acetylcholine.…”

“…Phosphatidylcholine is the main form of phosphoglycerides that contains the choline molecule as the head group. It represents the main phospholipid of the outer layer of the cellular and intracellular membranes of mammalian cells, and it accounts for 32.8% of the total glycerophospholipid content of the human brain (Javaid et al, 2021). The synthesis could be mediated by the direct methylation of the ethanolamine residue of phosphatidylethanolamine or via the Kennedy pathway.…”
