oxidants, CTAP III and NAP2 have already been described [17,18].Transferrin (TF) not only transports iron in all extracellular fluids but also exerts many additional properties, including cell growth stimulation, bacteriostatic action and inhibition of histamine release from mast cells and basophils [19][20][21][22][23]. TF is a 75 kDa glycoprotein containing 679 amino acids and two glycan chains. For each, the last residue is a sialic (N-acetyl neuraminic) acid which can be hydrolyzed by neuraminidase. Circulating TF mainly originates from hepatocytes, its intracellular precursor being asialoapotransferrin. Transferrin receptors, which are present throughout the body, are abundant (10 6 /cell) on the basophil plasma membrane [24]. They are also involved in the development of the nervous system [25].Because of the known relationships between TF and histamine release, we decided to investigate the histamine-releasing ability of this glycoprotein in the plasma of MO patients. Jean-Marie Launay François Tabuteau André PradalierAbstract Patients with migraine without aura (MO), either during or between attacks, present elevated histamine levels in platelet-poor plasma but normal whole blood histamine levels, compared with controls. This finding is usually interpreted as an increased histamine release from basophils due to unidentified histamine-releasing factors. Compared with 10 control plasma samples, each sample from 12 MO patients (5 during and 7 between in attacks) contained normal amounts of iron and immunologically reactive transferrin but decreased transferrin iron-binding capacity. As transferrin inhibits histamine release in vitro, such a functional abnormality, probably due to modifications of the transferrin glycan moiety (desialylated transferrin), may well account for the increased histamine release observed in MO patients. We suggest that glycanmodified transferrin may be related to migraine histamine-releasing factors.

Automated Multicapillary Electrophoresis for Analysis of Human Serum Proteins

Background:We evaluated a new, automated multicapillary zone electrophoresis (CE) instrument (Capillarys ® , 4.51 software version; Sebia) for human serum protein analysis. Methods: With the Capillarys ␤1-␤2؉® reagent set, proteins were separated at 7 kV for 4 min in 15.5 cm ؋ 25 m fused-silica capillaries (n ‫؍‬ 8) at 35.5°C in a pH 10 buffer with online detection at 200 nm. Serum samples with different electrophoretic patterns (n ‫؍‬ 265) or potential interference (n ‫؍‬ 69) were analyzed and compared with agarose gel electrophoresis (AGE; Hydrasys ® -Hyrys ® , Hydragel protein(e) 15/30 ® reagent set; Sebia). Results: CVs were <3.5% for albumin, <11% for ␣ 1 -globulin, <4.1% for ␣ 2 -globulin, <7.4% for ␤-globulin, and <5.8% for ␥-globulin (3 control levels); measured throughput was 60 samples/h. In patients without paraprotein (n ‫؍‬ 116), the median differences between CE and AGE were ؊5.4 g/L for albumin, 4.0 g/L for ␣ 1 -globulin, 0.7 g/L for ␣ 2 -globulin, 0.6 g/L for ␤-globulin (P <0.001 for all fractions), and ؊0.1 g/L for ␥-globulin (not significant). More samples had at least one ␥-migrating peak detected by CE (n ‫؍‬ 135 vs 130; paraprotein detection limit, ϳ0.5-0.7 g/L), but fewer were quantified (n ‫؍‬ 84 vs 91) because of ␥-to ␤-migration shifts. Background: Congenital disorders of glycosylation (CDG) are usually diagnosed by isoelectric focusing (IEF) of serum transferrin (Tf). The aim of this study was to evaluate capillary zone electrophoresis (CZE) as a diagnostic alternative to IEF. Methods: We performed 792 CZE analyses of Tf, using the CEofix TM -CDT (carbohydrate-deficient transferrin) assay. Peak identification was based on relative migration times (RMTs) to reduce migration variability. Results: Tf profiles comprised three main groups (A-C).Groups A and B were characterized by one or two dominant tetrasialo-Tf peaks, whereas group C showed a widely variable Tf isoform composition. Group A was composed of four subgroups: a major group with a typical Tf profile (considered as reference group), two minor groups with decreased or moderately increased trisialo-Tf isoform, and a group showing the presence of unknown compounds with RMTs similar to mono-and disialo-Tf. However, these compounds were absent on IEF. Group C contained all profiles from patients with confirmed as well as putative CDG. From the reference group, 99% confidence intervals were calculated for the RMTs of the Tf isoforms, and percentiles representing the Tf isoform distributions were defined. Conclusions: All patients with abnormal IEF results and confirmed CDG were identified by CZE; thus, this method can be used as a diagnostic alternative to IEF in a manner suitable for automation. Because whole serum is analyzed, it should be kept in mind that CZE profiles can show substances other than Tf.
