Dynamic 31P–MRSI using spiral spectroscopic imaging can map mitochondrial capacity in muscles of the human calf during plantar flexion exercise at 7 T

Abstract: Phosphorus MRSI (31P–MRSI) using a spiral‐trajectory readout at 7 T was developed for high temporal resolution mapping of the mitochondrial capacity of exercising human skeletal muscle. The sensitivity and localization accuracy of the method was investigated in phantoms. In vivo performance was assessed in 12 volunteers, who performed a plantar flexion exercise inside a whole‐body 7 T MR scanner using an MR‐compatible ergometer and a surface coil. In five volunteers the knee was flexed (~60°) to shift the major… Show more

“…Moreover, additional measurements with increased force and a flexed knee were attempted, but did not generate similar depletion to that in gastrocnemius medialis or were not well tolerated by the subject. The observation that the PCr depletion in soleus was stronger with a flexed knee than with a straight leg, but without depletion reaching the maximum depletion levels of gastrocnemius, has also been reported by Valkovic et al…”

“…A relation between knee angle and the recruitment pattern of gastrocnemius and soleus is expected according to anatomy and has been explored with 1 H MRI and 31 P MRS . The purpose of this study was to investigate the effect of knee angle on metabolic changes in those muscles in further detail, in order to establish a more comprehensive model by acquiring localized 31 P MRS data in exercising calf muscles with a simple and time‐efficient method with four different knee angles.…”

“…Recently, a constant-density spiral spectroscopic readout has been proposed for dynamic 31 P-MRSI experiments [261]. The use of temporal interleaved spirals allowed the acquisition of a 14 × 14 matrix in 10 s; thus, a simple measurement protocol using repetitive exercise can be used.…”

“…These processes have been investigated in several studies employing either 31 P MRS and MRI or 1 H arterial spin labeling (ASL) MRI in calf muscles during and after exercise. 1 H ASL is an approach to noninvasively assess local perfusion rates, while 31 P MRS is a well‐established method to investigate the energy metabolism in vivo.…”
