Dynamic Regulation of Myosin Light Chain Phosphorylation by Rho-kinase

Abstract: Myosin light chain (MLC) phosphorylation plays important roles in various cellular functions such as cellular morphogenesis, motility, and smooth muscle contraction. MLC phosphorylation is determined by the balance between activities of Rho-associated kinase (Rho-kinase) and myosin phosphatase. An impaired balance between Rho-kinase and myosin phosphatase activities induces the abnormal sustained phosphorylation of MLC, which contributes to the pathogenesis of certain vascular diseases, such as vasospasm and h… Show more

“…Rho-associated kinase (ROCK) is a known Rho effector protein and it may act to increase cytoskeletal tension by directly or indirectly facilitating the phosphorylation of myosin light chain [55]. To investigate if ROCK inhibition could stimulate a restoration in cell spreading, Y27632 and thiazovivin, two known ROCK inhibitors [56,57], were used to treat B16/GPR4 cells.…”

“…52, and for large barbed end density or the "coherent" regime, the protrusion rate is not sensitive to the number of barbed ends. We also assume that the Rho-activation leads to the activation of ROCK and myosin phosphorylation (53). We then propose that the increase in spine size is proportional to the increase in actin barbed ends (similar to the leading edge pushing velocity) and that the decrease is proportional to the amount of phosphorylated myosin.…”
