Dynamic cross-flow filtration: enhanced continuous small-scale solid-liquid separation

Abstract: In a previous study, a small-scale dynamic filtration device (SFD) was analyzed and the basic mechanisms governing the filtration process were characterized. The present work aims at improving the device's performance in terms of actual production. Various operation modes were tested in order to increase permeate flow and concentration factors (CF), while maintaining a fully continuous production mode. Both, a vacuum-enhanced and a pulsating operation mode, proved to be superior to the currently implemented op… Show more

“…As the demand for small-scale continuous isolation and drying technology is increasing, an effort was made in the past years to develop such equipment . As the requirements according to yield and quality differ extremely for small- and large-scale products, existing industrial continuous filtration processes cannot simply be scaled down.…”

“…Moreover, they may be subjected to thermal degradation or have an agglomeration or attrition tendency. To address these issues, a technique of drying at low temperatures while forcing gentle internal powder movement may be applied. As thermal drying is energy-intensive, moisture is removed mechanically as much as possible, typically via filtration. In this context, a technology that allows a continuous drying of slurries or powders with a high moisture content after filtration could be beneficial. Reducing the residence time in comparison to batch processing can help minimize the risk of thermal degradation .…”

“…Cross flow filtration is a well-known continuous filtration technique, mainly utilized during the processing of biological products to concentrate slurries. , Gursch et al. and Kossik et al reported studies related to this technique for API suspensions with a membrane-based device , and a rotary drum filter. , Researchers at MIT designed, built, and tested the prototype of a small-scale continuous pharmaceutical filtration instrument using linear motion . Pfizer developed a semicontinuous filter-drier with a 3-way valve, which filtered small amounts of slurry (15–30 mL) every cycle, yielding a kg per a day productivity .…”
