During stationary phase, Beijerinckia derxii shows nitrogenase activity concomitant with the release and accumulation of nitrogenated substances

Abstract: Beijerinckia derxii, a free-living nitrogen-fixing bacterium, maintained an increasing nitrogenase specific activity during the stationary growth phase. To verify the destination of the nitrogen fixed during this phase, intra and extracellular nitrogenated contents were analyzed. Organic nitrogen and amino acids were detected in the supernatant of the cultures. An increase in intracellular content of both nitrogen and protein occurred. Cytoplasmic granules indicated the presence of arginine. The ability of a n… Show more

“…Inoculation of rice seeds with Beijerinckia showed that this genus is able to multiply in the soil, establishing itself in large numbers and reducing the number of other competing microorganisms (Döbereiner and Ruschel 1961). It has been observed (Miyasaka et al 2003) that B. derxii maintains an increasing nitrogenase-specific activity during the stationary growth phase. To verify the destination of the nitrogen fixed during this phase, intra-and extracellular nitrogenated contents were analyzed.…”

“…This suggested that B. derxii contributes to the environment by both releasing nitrogenated substances and accumulating substances capable of being consumed after its death. Moreover, the ability of B. derxii to release certain plant growth regulators (indoleacetic acid, ethylene and polyamines) and amino acids into the growth medium has also been addressed (Thuler et al 2003). The production of those substances was compared using cultures in which nitrogenase was active (N-free medium) and cultures in which nitrogenase was repressed (combined-N cultures) and with and without agitation.…”

“…Because endophytic diazotrophs have only been observed in intercellular spaces, vascular tissue, aerenchyma and dead cells and not within living host cells, James [45] suggested that N transfer from these organisms is likely to be dependent on their death and release of fixed N. Transfer to plants of N fixed by diazotrophs or N contained in non-fixing microbial biomass in the soil or rhizosphere is also likely to be dependent on the death of these bacteria and release of ammonium or amino acids [151,152], although excretion of nitrogenous substances during bacterial growth can also occur (e.g. Beijerinckia derxii [153]; cyanobacteria [154]).…”

“…Esta bactéria é capaz de excretar grande variedade de aminoácidos assim como substâncias nitrogenadas reguladoras do crescimento vegetal (poliaminas e ácido 3-indol acético) ou não nitrogenadas [etileno] (THULER et al, 2003a). Também foi demonstrada a capacidade de armazenamento de substâncias nitrogenadas (MIYASAKA et al, 2003).…”
