Duplication of Subcytoband 11E2 of Chromosome 11 Is Regularly Associated with Accelerated Tumor Development in v-abl/myc-Induced Mouse Plasmacytomas

Abstract: Chromosome 11 aberrations constitute the second most frequent chromosomal aberration in mouse plasmacytomas (PCTs) in which both the myc and abl oncogenes are constitutively expressed. In these tumors, previous G-banding studies had revealed numerical aberrations including duplication of the entire chromosome 11 or segments of telomeric bands D and E. The trisomy of chromosome 11 was always associated with accelerated pristane + v-abl/myc-induced PCT development. In the present study, PCT development was studi… Show more

“…The plasmacytoma cell line MOPC460D, a gift of J. Mushinski (National Institutes of Health, Bethesda), were cultured as described (31). T38H mice (32) were obtained from Harwell, UK (33). The mice were kept under specific pathogen-free (SPF) conditions.…”

“…Two cell types were used, a PreB cell line of BALB/c origin and primary B cells from congenic [T38HxBALB/c]N wild‐type mice . Procedures were conducted according to Animal Protocol 11–019, approved by Central Animal Care Services, University of Manitoba (Winnipeg, MB, Canada).…”

“…This unique mouse model exhibits a long chromosome T(11;X) and a short chromosome T(X;11). The short chromosome T(X;11) contains cytoband 11E2 and parts of cytoband E1 ( Figure 1 ) [19] . To determine the chromosome orientation in cancer cells and in the same cell lineage, we studied mouse PCT induced in this unique mouse model.…”

“…There are slow- and fast-developing PCTs. Slow-onset PCTs are induced only by pristane (2,6,10,14-tetramethylpentadecane) [20] ; fast-onset PCTs are induced by pristane and v-abl/myc [19] , [21] . In the current study, we focused on the fast-onset PCTs.…”
