Duck Hepatitis A Virus Type 1 Induces eIF2α Phosphorylation-Dependent Cellular Translation Shutoff via PERK/GCN2

Abstract: Duck hepatitis A virus type 1 (DHAV-1) is one of the most deadly pathogens that endanger the duck industry. Most viruses usually turn off host translation after infection to facilitate viral replication and translation. For the first time report to our knowledge, DHAV-1 can induce eIF2α phosphorylation and inhibit cellular translation in duck embryo fibroblasts (DEFs). Moreover, the activity of DHAV-1 in the cells caused obvious eIF2α phosphorylation, which has nothing to do with the viral protein. Subsequentl… Show more

“…It is well known that protein is the material basis of life activities (63). The same protein exists in different structures and performs different biological functions due to post-translational modifications (64). Current studies have shown that the cargoes carried by MDSCs-Exos are mainly involved in the immunosuppressive effect of MDSCs (65).…”

“…IRES-mediated translation initiation of viral mRNAs differs from cap-dependent translation initiation of most eukaryotic mRNAs ( Leppek et al, 2018 ). Similar to other picornaviruses, DHAV-1 can overcome the massive cellular translation block after infection, and the mechanism driven by IRES provides advantages for viral protein synthesis ( Han et al, 2021 ; Liu Y. et al, 2021 ). The activity of IRESs depends on specific viral RNA motifs, with each type sharing a common RNA structural core ( Martinez-Salas et al, 2017 ).…”
