Dual-Wavelength Electrochemiluminescence Ratiometric Biosensor for NF-κB p50 Detection with Dimethylthiodiaminoterephthalate Fluorophore and Self-Assembled DNA Tetrahedron Nanostructures Probe

Abstract: In this work, we fabricated a dual-wavelength electrochemiluminescence ratiometric biosensor based on electrochemiluminescent resonance energy transfer (ECL-RET). In this biosensor, Au nanoparticle-loaded graphitic phase carbon nitride (Au-g-C3N4) as a donor and Au-modified dimethylthiodiaminoterephthalate (TAT) analogue (Au@TAT) as an acceptor were investigated for the first time. Besides, tetrahedron DNA probe was immobilized onto Au-g-C3N4 to improve the binding efficiency of the transcription factor and EC… Show more

“…Electrochemiluminescence (ECL) immunoassay, the most advanced labeling immunoassay technique, is a new generation of labeling immunoassay technique after radioimmunoassay, enzyme immunoassay, fluorescence immunoassay and chemiluminescence immunoassay, which is sensitive, rapid and stable, and is the technological leader in solid-phase labeling immunoassay [25] , [26] , [27] . Electrochemiluminescence is a specific chemiluminescence reaction triggered by electrochemistry on the electrode surface, which is actually a perfect combination of two processes: electrochemistry and chemiluminescence [2] , [28] , [29] , [30] . The main difference between electrochemiluminescence and ordinary chemiluminescence is that the former is an electrically initiated luminescence reaction, cyclic and multiple luminescence, while the latter is a single instantaneous luminescence reaction initiated by a mixture of compounds [31] , [32] .…”

“…The classically modified Randles circuit (inset in Figure 5 B) can simulate the electrochemical process of the modified electrode. The resistance of the electrode surface, the electron transfer and matter transfer during the redox process are mainly expressed through the resistance to charge transfer (Rct), which is precisely quantified especially through the Nyquist diagram’s high-frequency semicircle [19] . The impedance value of the bare electrode (curve a) was the smallest.…”

“…Au nanoparticles (AuNPs) have good biocompatibility and are widely used in the composite of functional nanomaterials [18] . Graphite-C 3 N 4 (Au-g-C 3 N 4 ) decorated with AuNPs has attracted attention as a donor material because of its good biocompatibility and excellent luminescence efficiency, but there is still a great lack of suitable acceptor materials [19] . Coincidentally, Ru(II) has a strong UV-vis absorption peak at 460 - 470 nm overlapping with the ECL emission peak of Au-g-C 3 N 4 .…”

“…We successfully synthesized Au-g-C 3 N 4 with reference to our previous work [ 16 ]. In simple terms, 40 μL of HAuCl 4 ·xH 2 O (0.01 M) solution and 4 mL g-C 3 N 4 ethanol-water dispersion (15 mg L −1 ) were mixed and sonicated for 10 min, followed by stirring at room temperature for 4 h to get a mixture solution of g-C 3 N 4 and HAuCl 4 .…”

“…On the other hand, electrochemiluminescence (ECL) based sensors have been rapidly developed due to their advantages of low background interference and high sensitivity [ 1 , 16 , 17 ].ECL sensors have shown potential for ultra-sensitive and real-time analysis. In the ECL process, a substance near the electrode is triggered by an electrochemical method to produce an excited state and generate light emissions.…”
