Dual RNA-seq identifies human mucosal immunity protein Mucin-13 as a hallmark of Plasmodium exoerythrocytic infection

Abstract: The exoerythrocytic stage of Plasmodium infection is a critical window for prophylactic intervention. Using genome-wide dual RNA sequencing of flow-sorted infected and uninfected hepatoma cells we show that the human mucosal immunity gene, mucin-13 (MUC13), is strongly upregulated during Plasmodium exoerythrocytic hepatic-stage infection. We confirm MUC13 transcript increases in hepatoma cell lines and primary hepatocytes. In immunofluorescence assays, host MUC13 protein expression distinguishes infected cells… Show more

“…PBANKA_0521700 is preferentially expressed in the ring stages of the IDC and is refractory to disruption in the BS (21,24), hampering functional studies of this gene. Our data, in conjunction with previously reported P. berghei RNA-Seq (8,21,24,25) and single-cell studies covering the entire life cycle (9), could be useful to refine hypotheses about the functions and targets of this TF, as well as other AP2 TFs.…”

“…Recent development of dual RNA-seq analysis has assisted studies designed to measure transcripts of both pathogen and host during infection (29)(30)(31)(32). Most dual RNA-seq studies have used host cell lines to analyze pathogen-host interactions (33)(34)(35)(36)(37)(38)(39)(40), resulting in new information about the interplay between the two. However, these experiments share the inherent limitation that cell culture infection models exclude the influence of many host factors and thus are limited compared to the intact-animal milieu.…”
