Dual regulation of Akt and glutathione caused by isoalantolactone effectively triggers human ovarian cancer cell apoptosis

Abstract: Ovarian cancer is one of leading causes of cancer death in gynecological tumor. Isoalantolactone (IL), present in several medicinal plants, exhibits various biological activities, and its mechanism underlying anti-ovarian cancer activity needs to be further investigated. Here, we find that IL inhibits the proliferation of SKOV-3 and OVCAR-3 cells by causing G2/M phase arrest and inducing apoptosis. Moreover, IL decreases intracellular glutathione (GSH) level, and induces reactive oxygen species (ROS) generatio… Show more

“…Ovarian cancer cell lines (SKOV-3 and OVCAR-3) treated with IAL underwent cell cycle arrest at G2/M phase arrest and apoptosis. IAL exerted its apoptotic effects on these cancer cell lines by increasing intracellular ROS, amplifying Bax, suppressing Bcl-2, and activating caspase 3 and PARP ( Xie et al, 2023 ). Two studies have been conducted to assess the effects of IAL on the proliferation and apoptosis in pancreatic cancer.…”

“…Intracellular glutathione (GSH) is an intracellular antioxidant that is present abundantly and regulates the redox balance in the cells 18 . Cancer cells have higher levels of GSH compared with their noncancerous counterparts that protects the cancer cells against oxidative damage and apoptosis caused by extrinsic factors 19 . Increased GSH significantly reduces oxidative damage and leads drug resistance in cancer cells 20 .…”

“…18 Cancer cells have higher levels of GSH compared with their noncancerous counterparts that protects the cancer cells against oxidative damage and apoptosis caused by extrinsic factors. 19 Increased GSH significantly reduces oxidative damage and leads drug resistance in cancer cells. 20 Moreover, GSH depletion causes more reactive oxygen species (ROS) production that activates the transcription of apoptosis-associated genes.…”
