Dual-energy micro-CT of the rodent lung

Abstract: The purpose of this work is to investigate the use of dual-energy microcomputed tomography (CT) for the estimation of vascular, tissue, and air fractions in rodent lungs using a postreconstruction three material decomposition method. Using simulations, we have estimated the accuracy limits of the decomposition for realistic micro-CT noise levels. Next, we performed experiments involving ex vivo lung imaging in which intact rat lungs were carefully removed from the thorax, injected with an iodine-based contrast… Show more

“…First, the choice of domain and range weighting functions used here was based on brute-force optimizations of BF using similar data as documented in (Clark et al , 2012). Second, we previously demonstrated the superiority of BF to Gaussian smoothing and median filtration using our grayscale CT data (Clark et al , 2012) and in DE applications (Badea et al , 2012). Here, building on the conclusions in these papers, joint BF is seen to outperform classic BF.…”

“…In previous studies, applying BF to each energy independently worked well as a preprocessing step for improving the precision and sensitivity of DE decomposition (Badea et al, 2012). Here, we compared this approach (“independent” BF) with a novel extension of BF designed to exploit spectral contrast (“joint” BF):…”

“…15 nm AuroVist from Nanoprobes, Inc.) have gained importance in preclinical imaging because of the aforementioned contrast improvement over iodine contrast agents. Previously, we have demonstrated many preclinical, functional imaging applications using dual-energy micro-CT involving the separation of iodine and calcium or iodine and gold, including classification of atherosclerotic plaque composition (Bhavane et al 2013), non-invasive measurement of lung (Badea et al 2012), lung tumor (Ashton et al 2014b), and myocardial perfusion (Ashton et al 2014a), and the classification of tumor aggressiveness and therapy response in primary sarcoma tumors (Clark et al 2013, Moding et al 2013). …”

“…In vivo high-resolution micro-CT allows for longitudinal image-based measurements in animal models of lung diseases, but it remains technically challenging because of respiratory and cardiac movement artifacts (3,6). Noninvasive, serial imaging of animal models should ideally result in quantitative datasets that allow for longitudinal assessment, comparisons between different groups, including the effect of therapeutic interventions, and detailed topographic information documenting the extent of disease in individual animals.…”
