Dual DNA topoisomerase 1 and tyrosyl‐DNA phosphodiesterase 1 inhibition for improved anticancer activity

Abstract: Tyrosyl‐DNA phosphodiesterase 1 (Tdp1) is a DNA repair enzyme that catalyzes the hydrolysis of the phosphodiester bond in the DNA‐topoisomerase 1 (Top1) covalent complex and repairs some other 3′‐end DNA adducts. Currently, Tdp1 functions as an important target in cancer drug design owing to its ability to break down various DNA adducts induced by chemotherapeutics. Tdp1 inhibitors may sensitize tumor cells to the action of Top1 poisons, thereby potentiating their effects. This mini‐review summarizes findings … Show more

“…However, TDP1 removes the stalled TOP1-DNA complex, reducing the efficacy of the drugs and leading to resistance [ 7 , 8 , 9 ]. As has been confirmed in gene knockout studies in mice and human cell lines, TDP1 contributes to the development of drug resistance in a number of cancers (see [ 3 ] and references therein). Indeed, human cell lines with mutant or missing TDP1 as well as TDP1-knockout mice are both hypersensitive to the well-established TOP1 inhibitor camptothecin and its derivatives.…”

“…Tyrosyl-DNA-phosphodiesterase 1 (TDP1) is a DNA repair enzyme and a promising therapeutic target to enhance established cancer treatment [ 1 , 2 , 3 , 4 ]. TDP1 plays a key role in the removal of DNA lesions, in particular those caused by DNA topoisomerase I (TOP1) inhibitors.…”

“…TDP1/2 are enzymes responsible for the reparation of DNA breaks induced by topoisomerase poisons [ 57 , 211 , 212 ]. TDP1/2 inhibitors are capable of improving cancer cells’ sensitivity to these poisons [ 213 ].…”

“…One of the current approaches to increase the efficacy of clinically-established antitumor therapy is the inhibition of DNA repair enzymes that counteract the effect of DNA-damaging chemotherapy agents [23][24][25]. One of these important enzymes is tyrosyl-DNA phosphodiesterase 1 (Tdp1) [26].…”
