Drugging the efferocytosis process: concepts and opportunities

Abstract: The daily removal of billions of apoptotic cells in the human body via the process of efferocytosis is essential for homeostasis. To allow for this continuous efferocytosis, rapid phenotypic changes occur in the phagocytes enabling them to engulf and digest the apoptotic cargo. In addition, efferocytosis is actively anti-inflammatory and promotes resolution. Owing to its ubiquitous nature and the sheer volume of cell turnover, efferocytosis is a point of vulnerability. Aberrations in efferocytosis are associat… Show more

“…This would prevent a necrotic response to a controlled, cell death in the body, however, in atherosclerosis this mechanism is altered. This efferocytosis impairment is responsible for the chronic inflammation in a number of inflammatory diseases, and there are extremely limited treatments currently available for this, however, the use of miR-33 could be a future therapeutic avenue targeting defective efferocytosis, due to the role of the abca1 gene in functional efferocytosis ( Mehrotra and Ravichandran, 2022 ).…”

“…This was unexpected since the transcriptional profile of Type 2 polarized eosinophils was shown to display marked overlap and similarity with that of M2-polarized macrophages ( 6 ). The different transcriptional phenotype of eosinophils and macrophages in response to IL-4 and apoptotic cells likely results from the fact that macrophages express a large and different repertoire of receptors and intracellular molecules that can bind, engulf and digest apoptotic cells ( 54 ).…”

“…We were the first in 2001 to propose the use of apoptotic cells as a cell-based therapy approach ( 42 ). However, this is out of the scope of this review (for recent reviews please refer to ( 21 , 24 , 43 ). Here, we want to focus on the contribution of macrophages to the resolution therapy.…”
