L’amélioration des méthodes contraceptives et la recherche de nouvelles approches innovantes, avec un bénéfice médical ajouté, permettraient de satisfaire plus de couples, aux différentes étapes de leur vie reproductive. Les systèmes de délivrance s’améliorent, visant à une utilisation autonome et à une longue durée d’action, ce qui en facilite ainsi l’observance. Auto-injections, patchs à microaiguilles, anneaux vaginaux multipores, qui délivrent des molécules contraceptives et des molécules anti-infectieuses, promettent ainsi une double prévention, la grossesse non désirée et les maladies sexuellement transmissibles. Enfin, de nouveaux systèmes intra-utérins et de contraception permanente sont à l’étude. La recherche de nouveaux contraceptifs est une nécessité et doit se poursuivre, afin de diminuer la mortalité maternelle liée aux grossesses multiples et aux avortements, qui restent souvent à risque dans beaucoup de pays.

The usage of 3D-printing for drug-eluting implants combines the advantages of a targeted local drug therapy over longer periods of time at the precise location of the disease with a manufacturing technique that easily allows modifications of the implant shape to comply with the individual needs of each patient. Research until now has been focused on several aspects of this topic such as 3D-printing with different materials or printing techniques to achieve implants with different shapes, mechanical properties or release profiles. This review is intended to provide an overview of the developments currently described in the literature. The topic is very multifaceted and several of the investigated aspects are not related to just one type of application. Consequently, this overview deals with the topic of 3D-printed drug-eluting implants in the application fields of stents and catheters, gynecological devices, devices for bone treatment and surgical screws, antitumoral devices and surgical meshes, as well as other devices with either simple or complex geometry. Overall, the current findings highlight the great potential of the manufacturing of drug-eluting implants via 3D-printing technology for advanced individualized medicine despite remaining challenges such as the regulatory approval of individualized implants.

Implantable drug delivery systems (IDDSs) play a vital role in treating chronic diseases by reducing dosing frequency and enhancing drug efficacy due to targeted delivery. In the present study, an IDDS was developed from electrospun cellulose acetate (CA) and polycaprolactone (PCL) nanofiber membranes. The implant core consists of a drug‐loaded CA nanofiber (CA + Vit.D3) enclosed in a rate limiting of the PCL membrane (CA + Vit.D3/PCL). The CA and PCL nanofibrous membranes were characterized using a scanning electron microscope (SEM), Fourier transform infrared spectroscopy, X‐ray diffraction, and UV–Vis spectroscopy. This research also investigated in‐vitro cytotoxicity and whether the PCL membrane prolonged drug delivery or led to enhanced mechanical properties. A smooth, beadless surface morphology was observed with fiber diameters of 325 ± 101 nm and 333 ± 79 nm for CA and PCL, respectively. In‐vitro drug release and tensile testing showed that surrounding the core's implants with a PCL membrane improved mechanical properties and kinetic drug release. The modulus and tensile strength of CA + Vit.D3/PCL were 161 ± 14 and 13.07 ± 2.5 MPa, respectively—these values were significantly higher than those obtained for CA + Vit.D3 (132 ± 52 MPa and 8.16 ± 2.36 MPa, respectively). The drug release pattern exhibited by CA + Vit.D3 was burst release, which fits the first‐order kinetic model. In contrast, CA + Vit.D3/PCL exhibited slow drug release, which fits the zero‐order kinetic model. In conclusion, based on the outcomes and facility of the technologies outlined in this article, electrospun CA and PCL nanofibers are suitable for developing long‐term IDDSs.
