Drug-adapted cancer cell lines as preclinical models of acquired resistance

Abstract: Acquired resistance formation limits the efficacy of anti-cancer therapies. Acquired and intrinsic resistance differ conceptually. Acquired resistance is the consequence of directed evolution, whereas intrinsic resistance depends on the (stochastic) presence of pre-existing resistance mechanisms. Preclinical model systems are needed to study acquired drug resistance because they enable: (1) in depth functional studies; (2) the investigation of non-standard treatments for a certain disease condition (which is n… Show more

“…Prostate cancer cell lines PC3, DU145, LNCaP, and 22Rv1 were obtained from the German Collection of Microorganisms and Cell Cultures (DSMZ). The DX-resistant sublines were derived from the Resistant Cancer Cell Line (RCCL) collection ( ) [ 73 ]. LNCaP cells were grown and subcultured in Iscove Basal medium (Biochrom GmbH, Berlin, Germany), and PC3, DU145, and 22Rv1 cells were grown in RPMI-1640 medium (Gibco, Thermo Fisher Scientific, Darmstadt, Germany).…”

“…The cell lines RT4, RT112, T24, and TCCSup were obtained from DSMZ. The cisplatin-resistant sublines were derived from the RCCL collection ( ) [ 37 ]. RT4 (grade 1), RT112 (grade 2/3), T24 (grade 3), and TCCSup (grade 4) represent transitional cell carcinomas.…”

“…The neuroblastoma cell line was established from a bone metastasis of a stage IV neuroblastoma patient 39 , in accordance with relevant guidelines and regulations. All drug-resistant sublines had been established by continuous exposure to stepwise increasing drug concentrations as previously described 12 , 39 and were derived from the Resistant Cancer Cell Line (RCCL) collection 7 . The cisplatin-resistant ovarian cancer sublines had been adapted to 1 µg/mL (COLO-704 r CDDP 1000 ) or 2 µg/ml (EFO-27 r CDDP 2000 and EFO-21 r CDDP 2000 ) cisplatin.…”

“…Such methods may also enable the discrimination between cell lines of the same genetic origin, a task for which established methods are currently lacking. Such approaches have the potential to improve the reliability of research results due to the reduction of the use of misidentified cell lines [4][5][6][7] . By using deep learning, we have demonstrated that, it is possible to authenticate cancer cell lines, including parental cancer cell lines and their drug-adapted sublines, based on image recognition.…”
