Saffron (Crocus sativus L.) is a commercial spice crop well-known throughout the world, valued for culinary, colorant, and pharmaceutical purposes. In China, Fusarium nirenbergiae was detected as causative agent of saffron corm rot, the most pervasive disease for the first time in 2020. In the present study, 261 Fusarium-like isolates were recovered from 120 rotted corms in four saffron producing fields at Zhejiang, Shanghai, and Yunnan provinces, China, in 2021. A combination of morpho-cultural features and multilocus sequence analysis (MLSA) of the concatenated rpb2 (DNA-directed RNA polymerase II largest subunit) and tef1 (translation elongation factor 1-α) partial sequences showed that the isolates from saffron belong to Fusarium nirenbergiae as well as F. commune, and F. annulatum with isolation frequencies of 58.2%, 26.8%, and 14.9%, respectively. Notably, F. commune was more prevalent than F. annulatum in the collected samples. Pathogenicity tests confirmed that both species were pathogenic on saffron corm. This is the first report of F. annulatum and F. commune causing corm rot of saffron, globally. Outcomes of the current research demonstrate that Fusarium spp. associated with saffron corm rot are more diverse than previously reported. Furthermore, some plants were infected by two or more Fusarium species. Our findings broaden knowledge about Fusarium spp. that inflict corm rot and assist the development of control measures.

Black yeasts have been isolated from acidic, low water activity, and thermally processed foods as well as from surfaces in food manufacturing plants. The genomic basis for their relative tolerance to food-relevant environmental stresses has not been well defined. In this study, we performed whole genome sequencing (WGS) on seven black yeast strains including Aureobasidium (n=5) and Exophiala (n=2) which were isolated from food or food production environments. These strains were previously characterized for their tolerance to heat, hyperosmotic pressure, high pressure processing, hypochlorite sanitizers, and ultraviolet light. Based on the WGS data, three of the strains previously identified as A. pullulans were reassigned as A. melanogenum. Both haploid and diploid A. melanogenum strains were identified in this collection. Single-locus phylogenies based on beta tubulin, RNA polymerase II, or translation elongation factor protein sequences were compared to the phylogeny produced through SNP analysis, revealing that duplication of the fungal genome in diploid strains complicates the use of single-locus phylogenetics. There was not a strong association between phylogeny and either environmental source or stress tolerance phenotype, nor were trends in the copy numbers of stress-related genes associated with extremotolerance within this collection. While there were obvious differences between the genera, the heterogenous distribution of stress tolerance phenotypes and genotypes suggests that food-relevant black yeasts may be ubiquitous rather than specialists associated with particular ecological niches. However, further evaluation of additional strains and the potential impact of gene sequence modification is necessary to confirm these findings.

The search for potential signs of Martian life has been planned and implemented by worldwide countries for decades. Due to the high expenditure on Mars exploration, more easily accessible Marslike regions on Earth are invaluable targets for astrobiology research to understand the detection of potential Martian biosignatures. In order to detect potential life signals beyond Earth, questions on how to define life and biosignatures need to be carefully inspected. This review summarizes scientific instrumental techniques, our "eyes" and "hands", that facilitate identifying and quantifying biosignatures on Mars. Scientific devices that can be applied in astrobiology include electromagnetic spectrum-based spectrometers, mass spectrometers, redox potential indicators, circular dichroism polarimeters, in situ nucleic acid sequencers, life isolation/cultivation systems, and imagers. These techniques should be first tested in Mars analog extreme environments on Earth to validate their practicality on Mars. To better understand the instrumental detectability of biosignatures on Mars through its evolutionary history, terrestrial Mars analogs are divided into four major categories according to their similarities to different geological ages of Mars (the Early-Middle Noachian Period, the Late Noachian-Early Hesperian Period, the Late Hesperian-Early Amazonian Period, and the Middle-Late Amazonian Period). Future missions are suggested to explore more on the early terrains in Mars' Southern Hemisphere once the landing issue is solved engineeringly, since these explorations permit investigating a continuum of the habitability shift through Mars geological history. Moreover, practical applications of scientific instruments listed above are briefly reviewed based on the four categories of Mars analogs, and instruments applied for autonomous robotic rover tests in Mars analogs are further discussed. For engineering efficiency, a Mars rover ought to be equipped with as few assemblable instruments as possible. Therefore, once candidate landing regions 1 Biosignatures in Mars analogson Mars are defined, the portable suites of instruments should be smartly devised on the basis of the geological, geochemical, geomorphological, and chronological characteristics of the landing regions. Laboratory-based experiments without engineering restrictions are further encouraging and appealing if Mars sample-return missions are successfully completed. To exclude false positive and false negative conclusions in life discovery, the combined use and replication studies of multiple analytical techniques must be performed to confirm the observational results.
