Draft Genome Sequence of Streptomyces clavuligerus NRRL 3585, a Producer of Diverse Secondary Metabolites

Abstract: Streptomyces clavuligerus is an important industrial strain that produces a number of antibiotics, including clavulanic acid and cephamycin C. A high-quality draft genome sequence of the S. clavuligerus NRRL 3585 strain was produced by employing a hybrid approach that involved Sanger sequencing, Roche/454 pyrosequencing, optical mapping, and partial finishing. Its genome, comprising four linear replicons, one chromosome, and four plasmids, carries numerous sets of genes involved in the biosynthesis of secondar… Show more

“…Recent fullgenome sequencing has validated those analyses and shown that the paralogue cluster is located on a linear megaplasmid while the clavulanic acid and clavam clusters are separated on the chromosome by more than 1 Mb, an unusual arrangement in view of the usual clustering of secondary metabolite biosynthetic genes [46,68].…”

“…Assembly and contig editing were performed with CLC Genomics Workbench (CLC bio, Inc.) and Phred/ Phrap/Consed. A total of 1,945 coding sequences (89.4% coding density) were predicted by Glimmer 3.02 and annotated using the information from GenBank, UniProt, COG, KEGG, Pfam, and RAST (21).…”

“…Since sequencing of the S. clavuligerus genome sequence has recently been completed (9,32), microarray-type analyses comparing transcription patterns of a ⌬ceaS2-IF mutant grown in liquid and solid medium may help to identify additional genes that are involved and provide greater insights into this complex regulatory system.…”
