Draft Genome Sequence of an Acinetobacter Genomic Species 3 Strain Harboring a blaNDM-1 Gene

Abstract: Here we report the draft genome sequence of one Acinetobacter genomic species 3 strain, D499, which harbors the bla NDM-1 gene. The total length of the assembled genome is 4,103,824 bp, and 3,896 coding sequences (CDSs) were predicted within the genome. A previously unreported bla NDM-1 -bearing plasmid was identified in this strain.

“…In contrast to most other countries where NDM has been mainly identified in Enterobacteriaceae , in China to date NDM has only been reported in Acinetobacter [9], [11], [12], [24] and Enterococcus faecium [13]. However, other types of MBLs, such as IMP, and other classes of acquired carbapenemases, such a KPC, have been reported in Enterobacteriaceae in China[25][29], but isolates of NDM-1-producing E.coli have not previously been identified in China.…”

“…Although nosocomial transmission of NDM-1 has occurred in many countries[26], [32], [33], medical tourism plays a significant role in the spread of NDM-1[34], and traveling to the Indian subcontinent is a significant risk factor of infection with an NDM-1-producing bacterium[35]. Patients in China who were found to have bla NDM-1 -carrying bacteria had no history of traveling to the Indian subcontinent or another country[9], [11], [13], [24]. In this study, the patient carrying the bla NDM-1 -positive E. coli strain had no foreign travel history.…”

“…However, the situation appears more complex, as in pNDM-BJ01 from Acinetobacter lwoffii (5), ISAba125 upstream of bla NDM-1 appears to be part of an ISAba125-mediated composite transposon (Tn125), flanked by 3-bp direct repeats (DRs), that has been inserted downstream of aphA6 in the context found in Tn5393d from Alcaligenes faecalis (6). In the closely related plasmids pNDM-BJ02 from A. lwoffii (5) and pAB_D499 from A. pittii (2), only a small fragment of ISAba125 remains downstream of bla NDM-1 , possibly due to recombination between two copies of a repeated short sequence. Tn125 flanked by different DRs has also been found on the A. baumannii chromosome (15,16).…”

“…Our isolate was related to only one other A. pittii strain, TG6411, but with a lower score of 221. A total of 13 A. pittii draft genomes have been described in the BioProject (http://www.ncbi.nlm.nih.gov/bioproject/); however, draft ge-nomes of only three isolates were published, including one draft genome of an NDM-1-producing A. pittii strain from China (13).…”
