Draft Genome Sequence of a Thermophilic Desulfurization Bacterium, Geobacillus thermoglucosidasius Strain W-2

Abstract: Geobacillus thermoglucosidasius strain W-2 is a thermophilic bacterium isolated from a deep-subsurface oil reservoir in northern China, which is capable of degrading organosulfur compounds. Here, we report the draft genome sequence of G. thermoglucosidasius strain W-2, which may help to elucidate the genetic basis of biodegradation of organosulfur pollutants under heated conditions.

“…One 3-demethylubiquinone-9 3-methyltransferase gene was also identified in this genome, which may be responsible for the metabolism of aromatics. This strain has potential for biodesulfurization and biodenitrification, which is similar to what is found with the previously reported strain Geobacillus thermoglucosidasius W-2 (6), based on genetic annotation. Moreover, enzymes from thermophiles are active and stable under extreme temperatures, broad pH ranges, and the presence of organic solvents, which may provide many different advantages from an industrial perspective (10).…”

“…Genome sequencing of A. pallidus W-12 was carried out using the Illumina HiSeq 2500 platform at Majorbio Bio-Pharm Technology Co. Ltd. (Shanghai, China). De novo assembly was performed by using SOAP denovo (version 2.04), followed by local hole filling and base correction of assembly results using GapCloser (version 1.12) (6). The reads were quality controlled by G+C deviation and base error rate.…”

“…However, the ANI between P. thermoglucosidasius W-2 and other P. thermoglucosidasius strains is about 92.17%, slightly lower than the cutoff score 95% of defining same species [24]. It might be caused by the low-quality sequence of the draft genome [25]. For Geobacillus strains, the recN gene is a more robust marker for identifying a new strain at the genus and species levels [11].…”
