Draft genome and description of Consotaella salsifontis gen. nov. sp. nov., a halophilic, free-living, nitrogen-fixing alphaproteobacterium isolated from an ancient terrestrial saline spring

Abstract: A free-living, nitrogen-fixing, mesophilic and facultative aerobe, designated strain USBA 369, was isolated from a terrestrial saline spring of the Colombian Andes. The non-sporulating rods (1.5×0.8 µm) with rounded ends stained Gram-negative and were motile by means of lophotrichous flagella. The strain grew optimally at 30 °C, at pH 6.9-7.5 and with 1.5 % (w/v) NaCl. The major fatty acids detected were C18 : 1ω7c and C19 : 0 cyclo ω8c, and the respiratory lipoquinone ubiquinone 10 (Q-10) was present. The gen… Show more

“…It is well known that a large subgroup of α-Proteobacteria has a non-canonical tRNA His lacking the G-1:C73 pair and has a non-canonical HisRS species that recognizes the non-canonical G1:U72 pair and A73 in addition to the GUG anticodon [ 17 , 57 , 58 , 59 , 60 ]. The two α-proteobacterial species are Afifella pfennigii DSM 17,143 ( Ap ) [ 17 ] and Consotaella salsifontis USBA 369 ( Cs ) [ 61 ], because the well-studied α-proteobacterial Caulobacter crescentus HisRS was not active enough in E. coli [ 60 ]. Two kinds of allo-tRNA chassis, S072 and S005, were engineered to have a typical α-proteobacterial tRNA His anticodon arm as their V-arms, to develop H072-αψHis-3 and αH005-αψHis-3/4/5 variants ( Figure 7 A).…”

“…The major named fatty acids are usually C 18 : 1 ω7 c and C 19 : 0 cyclo ω 8 c [2]. Members of the family Aurantimonadaceae have been isolated from various environments such as offshore surface seawater [3], deep-sea sediment [4], water-cooling system [5], surface-sterilized root of a halophyte [6], terrestrial saline spring [7], deep-sea hydrothermal vent plume [8] and poplar tree bark [9]. This study focuses on the description of a novel genus and species with the type strain R10 T was isolated from Antarctic gravel soil.…”

“…The morphology and motility of strains USBA 874 and USBA 960 T were determined by phase contrast microscopy (Eclipse 50i, Nikon) and by electron microscopy (transmission electron microscopy and scanning electron microscopy) for strain USBA 960 T . Negative staining, preparation of thin sections for electron microscopy and electron microscope examination were performed as described by Diaz-Cárdenas et al [20]. Cells of strains USBA 874 and USBA 960 T occurred singly or in pairs and were straight, short bacilli measuring 0.9-1.5 µm long and 0.3-0.6 µm wide with rounded ends and stained Gram-negative ( Fig.…”
