Doxycycline protects against ROS-induced mitochondrial fragmentation and ISO-induced heart failure

Abstract: In addition to their anti-bacterial action, tetracyclines also have complex biological effects, including the modification of mitochondrial protein synthesis, metabolism and gene-expression. Long-term clinical studies have been performed using tetracyclines, without significant side effects. Previous studies demonstrated that doxycycline (DOX), a major tetracyclin antibiotic, exerted a protective effect in animal models of heart failure; however, its exact molecular mechanism is still unknown. Here, we provide… Show more

“…Atria and great vessels were trimmed from the ventricles; the weight of the ventricles was measured and normalized to body mass. Afterward, ventricles were fixed in 10% formalin for histology or freeze clamped for Western blot analysis [ 16 ].…”

“…Left ventricular (LV), ejection fraction (EF), LV end-diastolic volume, LV end-systolic volume, and the thickness of the septum and posterior wall were determined. EF (percentage) was calculated by 100 × [(LVEDV−LVESV)/LVEDV] [ 16 , 17 ].…”

“…In other cells types, necroptosis activation can enhance aerobic respiration and ROS production (Yang et al, 2018). As ROS can trigger mitochondrial fragmentation by diverse mechanisms, including Drp-1 activation (Abuarab et al, 2017;Riba et al, 2017), necroptosis-dependent mitochondrial fragmentation could represent an executioner mechanism for axonal degeneration after injury.…”

“…It has yet to be demonstrated whether or not these new agents will be safe and can help to overcome the poor survival and high rates of early death in this highrisk group of patients. Also doxycycline, which may have a cardioprotective effect and may lead to reduced fibril formation, should be studied for its clinical potential in this high-risk population [37][38][39]. Finally, early diagnosis of the disease remains crucial, since early treatment is associated with better survival outcomes.…”
