This study assessed the in vitro antibacterial activity of combinations of amikacin and doxycycline or tigecycline against multidrug-resistant E. coli isolates. Twenty-four different pulsotypes, including 10 extended-spectrum β-lactamase (ESBL)-, 10 carbapenem-resistant, 2 New Delhi Metallo-beta-lactamase (NDM)- and 2 Klebsiella pneumoniae carbapenemase (KPC)-E. coli isolates were collected. All 24 isolates were susceptible to amikacin and tigecycline. Only 30% of ESBL and 50% of carbapenem-resistant E. coli were susceptible to doxycycline. Both of the NDM-E. coli had a MIC of 64 μg/ml. The checkerboard method showed that for the ESBL- and carbapenem-resistant E. coli, the synergistic effects of amikacin/doxycycline were 80% and 90%, respectively. For the two KPC- and two NDM-E. coli, the FIC index of amikacin/doxycycline were 0.5/0.375 and 0.5/0.281, respectively. For the ESBL- and carbapenem-resistant E. coli isolates, the combinations of amikacin and doxycycline exhibited synergistic activities against 80%, and 80% and 10% vs 60%, and 80% and 10% of the isolates at concentrations of 1x, 1/2x and 1/4xMIC, respectively. The synergistic effect seems to be similar for doxycycline and tigecycline based combinations with amikacin. In conclusion, the antibacterial activity of doxycycline can be enhanced by the addition of amikacin and is observed against most multidrug-resistant E. coli isolates.

For the past two years, doxycycline has been employed hugely for the treatment of COVID 19 over the globe. Excessive use of doxycycline can result in bacteria and gene resistance, which affects the future treatment of infectious diseases. Furthermore, unused doxycycline left from the hospital and pharmaceutical industries may have an adverse effect on the environment, posing a significant menace to modern society. As a result, doxycycline detection is required. Herein, we developed blue luminous nitrogen-doped carbon quantum dots (N-CQDs) using ascorbic acid and diethylenetriamine (DETA) as carbon and nitrogen sources via a microwave-assisted technique for the differential detection of doxycycline (DC) via a fluorescence quenching mechanism, even when other tetracycline derivatives interfere. The quenching mechanism has been elaborately explained by using a Stern−Volmer plot, UV−vis and fluorescence spectroscopy, and TCSPC to attribute the static quenching and inner filter effect. In addition, the limit of detection of our suggested sensor is 0.25 μM. To confirm the structural properties and the size of the N-CQDs, FT-IR, Raman spectroscopy, HRTEM, DLS, and EDX have been performed. Moreover, this approach was used to identify doxycycline in pharmaceutical waste and bacterial cells. Because of its great sensitivity and selectivity, N-CQDs are ideal for measuring DC in environmental applications.

The active pharmaceutical ingredient (API) doxycycline (DOX) is a broad-spectrum antibiotic mainly used in the treatment of respiratory and urinary tract infections and, like many drugs, its efficacy may be affected by the crystal form. Up to now, only the crystal structure of doxycycline hyclate (DOX$HYC) (generic name of brand names such as DORYXÒ, PERIOSTATÒ, ATRIDOXÒ, and VIBRAMYCINÒ) has been reported. This study presents the single-crystal X-ray diffractometry structural characterisation of another crystal form, doxycycline monohydrate (DOX$H 2 O) (generic name of brand names such as MONODOXÒ and ORACEAÒ). The DOX$H 2 O structure was compared with the known DOX$HYC one in terms of intra-and intermolecular geometries, and their melting temperature, water solubility and dissolution rate were measured. These data allowed us to establish relationships between solid state properties related to the pharmaceutical performance of the two DOX crystal variants and their supramolecular structures for the first time. Both hyclate and monohydrate forms crystallise the DOX molecules as zwitterions in which their dimethylamine groups are protonated and one of their hydroxyl groups is deprotonated. Whereas two conformers were observed in the DOX$HYC (i.e., the amine group is next to the enolate in one of them (T1) and beside the carbonyl in the other one (T2)), only one (T2) was found in DOX$H 2 O. Additionally, in the hyclate form, the presence of ethanol in the crystal lattice could be related to a rotation around the C-C bond of the amide group, directing the oxygen toward the amine group in one (T1) of the two conformers present in this solid state phase. Meanwhile, in the other crystallographically independent molecule (T2), the amide nitrogen is on the same side as the amine. However, only the conformer similar to T1 in DOX$HYC was observed in DOX$H 2 O. The crystal packing of DOX$H 2 O was stabilised by several intermolecular hydrogen bonds, with each drug entity interacting with another two DOX and three water molecules in such a way that a compact supramolecular network was formed. This structure was saturated in terms of hydrogen bonding, which could be related to its lower solubility and dissolution rate relative to DOX$HYC.
