Downregulation of RNF128 activates Wnt/β-catenin signaling to induce cellular EMT and stemness via CD44 and CTTN ubiquitination in melanoma

Abstract: Background Ring finger proteins (RNFs) were involved in carcinogenesis. Here, we aimed to explore the detailed mechanism of RNF128 in the progression of melanoma. Methods We reanalyzed several gene expression profiles from the Gene Expression Omnibus (GEO) database and obtained the overlapped differential expressed RNF genes. Among them, RNF128 was selected to further explore its expression, the biological significance, and the underlying molecular mechanism, as well as… Show more

“…After 6 h, slides were treated with secondary antibodies and colored by DAB peroxidase substrate kit for IHC (Yeasen, China). The staining levels were quantitated with Image-Pro Plus 7.0 (Media Cybernetics, USA) as previously described [10]. In brief, three representative areas of each patient tissue were photographed (200×) and their density of positive staining were counted with the integrated absorbance and image area.…”

“…Subsequently, we interrogated the regulatory mechanism of DOCK9-AS2 in PTC. Wnt/β-catenin is a renowned pathway that regulates cell proliferation, metastasis, and stemness in cancer 30 , 31 . Therefore, we wondered whether DOCK9-AS2 influenced Wnt/β-catenin signaling.…”

“…However, since there are many downstream effectors of the PI3K/Akt pathway, we referred to the previous studies showing that Akt can phosphorylate GSK3β which sequentially increases β‐catenin accumulation in nucleus and thus β‐catenin activity 17 . Additionally, β‐catenin accumulation in nucleus has been confirmed to enhance the stemness in various cancers, 7,16,29 thus, we continued to investigate whether β‐catenin acts as the downstream effector of the PI3K/Akt pathway, and mediates the AGK‐inducing effects on NPC cell stemness. To confirm this issue, we firstly revealed that AGK positively regulated β‐catenin accumulation in nucleus, and thus β‐catenin activity.…”
