Downregulation of LSD1 suppresses the proliferation, tumorigenicity and invasion of papillary thyroid carcinoma K1 cells

Abstract: Abstract. The present study aimed to evaluate the effects of lysine-specific demethylase 1 (LSD1) downregulation, induced by small interfering RNA (siRNA) transfection, on the proliferation, colony formation, migration and invasion of the papillary thyroid carcinoma K1 cell line. The siRNA targeting LSD1 and scrambled non-targeting siRNA were each transfected into papillary thyroid carcinoma K1 cells. Downregulation of LSD1 mRNA and protein level was evaluated by reverse transcription-quantitative polymerase c… Show more

“…For example, microRNA-126 was found to be down-regulated in PTC tissues and cell lines, while its up-regulation could suppress cell proliferation, migration and invasion, and promote the apoptosis of PTC cells [28]. Up-regulated expression of lysine-specific demethylase 1 ( LSD1 ) has been detected in PTC cells, and its knockdown inhibited cell proliferation, tumorigenicity and invasion [29]. The growth and migration of PTC cells have been enhanced by the overexpression of hematopoietic pre-B cell leukemia transcription factor (PBX)-interacting protein ( HPIP ) [30].…”

“…Elevated levels of LSD1 in a wide spectrum of malignancies including prostate cancer ( 12 ), colon cancer ( 17 ), lung cancer ( 18 ), liver cancer ( 19 ), and thyroid carcinoma ( 20 ) are highly correlated with lymph node metastasis and poor prognosis. Based on our finding that elevated levels of LSD1 were highly related to the grades of the bladder cancers ( 16 ), we hypothesized that LSD1 plays an oncogenic role in BCa development and/or progression.…”

“…They showed that the PTC tissues had higher KDM1A expression than the paracancerous tissues. KDM1A down‐regulation can also suppress the invasive ability of PTC cell line K1, but how KDM1A affects PTC cells remains unknown.…”
