Downregulation of FOXO1 mRNA levels predicts treatment failure in patients with endometrial pathology conservatively managed with progestin-containing intrauterine devices

Abstract: Objective To examine hormone receptor expression levels and downstream gene activation in pre-treatment and post-treatment biopsies in a cohort of patients with endometrial pathology who were being conservatively managed with a progestin-containing intrauterine device (IUD). A molecular signature of treatment failure is proposed. Methods A retrospective analysis of pre- and post-treatment biopsy specimens from 10 women treated with progestin-containing IUD for complex atypical hyperplasia (CAH) or grade 1 en… Show more

“…Forkhead transcription factor 1 (FOXO1) is a member of the FOX transcription factor family which regulates a variety of important cellular processes including metabolism, cellular differentiation, apoptosis, and cell cycle progression . FOXO1 expression is associated with endometrial cancer progression in patients receiving progestin therapy . FOXO1 overexpression inhibits endometrial cancer cell proliferation, migration, and invasion .…”

“…Regarding risk of bias within study assessment, for the “Patient selection” domain, three studies were considered to be low risk of bias and 10 were considered unclear risk (because they did clearly not state that patients were consecutive and/or that only cancers with initial myometrial invasion were excluded).…”
