Downregulation of circRNA DMNT3B contributes to diabetic retinal vascular dysfunction through targeting miR-20b-5p and BAMBI

Abstract: Background: Diabetic retinopathy, a vascular complication of diabetes mellitus, is the leading cause of visual impairment and blindness. circRNAs act as competing endogenous RNA, sponging target miRNA and thus influencing mRNA expression in vascular diseases. We investigated whether and how circDNMT3B is involved in retinal vascular dysfunction under diabetic conditions. Methods: qRT-PCR was performed to detect expression of circDNMT3B, miR-20b-5p, and BAMBI in retinal microvascular endothelial cells under dia… Show more

“…Emerging evidence has revealed that circRNAs act as miRNA sponges in various human diseases, including diabetic retinal vascular dysfunction and myocardial fibrosis (26,27). The results of the present study demonstrated that miR-370-3p was a target of circ_0010283.…”

“…Circular RNAs (circRNAs) are endogenous RNAs produced by back‐splicing and have neither a polyadenylated tail nor a 5′ to 3′ polarity 3,4 . With the development of next‐generation sequencing and bioinformatics methods, many circRNAs have been shown to be involved in the pathogenesis of various diseases such as diabetes, heart diseases, psychiatric diseases, and cancers 5‐8 …”

“…31 Recent study also reveals that miR-20b-5p is sponged by circRNA DNA Methyltransferase 3 Beta (DMNT3B) in human retinal microvascular endothelial cell. 32 In addition, let-7e-5p overexpression inhibited cell proliferation, wound healing, migration, and invasion in head and neck squamous cell carcinoma cells. 33 Chiam et al found that circulating serum exosomal let-7e-5p may be a useful marker to distinguish esophageal adenocarcinoma from controls and Barrett's esophagus.…”
