Down‐regulation of TIMP2 by HIF‐1α/miR‐210/HIF‐3α regulatory feedback circuit enhances cancer metastasis in hepatocellular carcinoma

Abstract: Cancer metastasis is a multistep process that involves a series of tumor‐stromal interaction, including extracellular matrix (ECM) remodeling, which requires a concerted action of multiple proteolytic enzymes and their endogenous inhibitors. This study investigated the role of tissue inhibitor of metalloproteinases (TIMP) 2 in the context of hepatocellular carcinoma (HCC) metastasis. We found that TIMP2 was the most significantly down‐regulated member among the TIMP family in human HCCs. Moreover, TIMP2 undere… Show more

“…HIF-3a1 is reported to promote colorectal cancer cell growth through noncanonical, transcription-independent activation of the JAK-STAT3 signaling pathway (27). Ectopic overexpression of a HIF-3a gene lacking the 3 0 untranslated region might disturb the regulatory circuit by altering HIF-1a transcriptional activity (28).…”

“…The prognosis of HCC patients remains unsatisfactory due to a high incidence of metastasis, which contributes to around 90% of cancer-related mortality [42, 43]. Therefore, it is necessary to identify prognostic markers and treatment targets for metastatic HCC.…”

“…As both HIF-1α and HIF-2α play important roles in promoting HCC progression, the combined inhibition of HIF-1α and HIF-2α can synergistically inhibit HCC development and improve patient prognosis (44)(45)(46)(47). Recently, sorafenib was reported to decrease HIF-1α synthesis and enhance HIF-2α synthesis (10).…”
