Down‐regulation of lncRNA HCG11 promotes cell proliferation of oral squamous cell carcinoma through sponging miR‐455‐5p

Abstract: Background As a type of head and neck squamous cell carcinoma (HNSCC), oral squamous cell carcinoma (OSCC) has a high incidence and low survival rate. Frequent deletion of protein tyrosine phosphatase receptor type sigma (PTPRS) has been found in HNSCC. Long non‐coding RNA (lncRNA) HCG11 and miR‐455‐5p have been reported to be involved in several cancers, in which miR‐455‐5p was found to be up‐regulated in the OSCC. However, the role of HCG11 in OSCC development is still unclear. Methods Several co‐transfectio… Show more

“…Recently, an increasing number of studies have been devoted to clarifying the pivotal roles of lncRNAs in cancers, including OS. With regard to HCG11, it has been identified to be aberrantly expressed in some cancers and function as an oncogene or a tumor suppressor gene [ 12 – 19 ], not including OS yet. Consistent with most of the tumors reported, our data suggest that HCG11 is downregulated and exerts a tumor suppressive role in OS.…”

“…HCG11 has been verified to act as a tumor suppressor gene in many cancers, including oral squamous cell carcinoma, non-small cell lung cancer, cervical cancer, laryngeal carcinoma, glioma and prostate cancer [ 12 17 ]. However, in some cancers, such as hepatocellular carcinoma and gastric cancer, HCG11 has been validated to be an oncogene [ 18 , 19 ].…”

“…Aiming at investigating the molecular mechanism of anti-tumor activity for LINC00472 in OSCC patients, the potential miRNA targets were predicted to find that LINC00472 has a binding site for miR-455-3p ( Figure 3a ). Then, miR-455-3p was selected for further experimentation, as it ranked first among all potential targets and multiple explorations showed a pro-tumor function of OSCC miR-455-3p [ 28 , 29 ]. To confirm the regulatory role of LINC00472 on miR-455-3p, a dual-luciferase reporter assay was conducted.…”

“…Furthermore, hsa-miR-455-5p isolated from plasma has been proved to be an early diagnostic indicator of OSCC [41]. It may regulate the OSCC cell proliferation and promote the OSCC tumorigenesis through affecting PTPRS related pathway and transforming growth factor-β (TGF-β) pathway, these pathways were also common in HNSCC [42]. Each of these miRNAs seems to have a certain impact on tumorigenesis, and combining these miRNAs into a feature can more effectively predict cancer outcomes.…”
