Dose response and repair kinetics of γ-H2AX foci induced by in vitro irradiation of whole blood and T-lymphocytes with X- and γ-radiation

Abstract: gamma-H2AX foci response and repair kinetics depend on irradiation conditions and radiation quality, possibly linked to Bystander response.

“…Therefore, the use of gH2AX foci loss as a biomarker of susceptibility in studies investigating the effects of less than 10 mGy might not be suitable because the kinetics of foci loss will be difficult to establish. It should be also noted that a similar level of persistent radiation induced gH2AX foci, depending on radiation quality (X rays vs. grays) and types of sample (whole blood vs. isolated lymphocytes), was reported after exposure to 5 and 200 mGy [264]. Regarding the use of gH2AX foci frequency as a biomarker of very low dose exposure, the persistence of the foci might be an opportunity to increase the time frame of sampling and testing after irradiation and also to discriminate between irradiated and non irradiated cells.…”

“…Both the observations that different radiation qualities yield: (a) Different numbers of foci per unit dose (see also Depuydt et al 2013, Beels et al 2010, and (b) differences in foci disappearance necessitate the establishment of distinctive dose-effect curves at different time-points as previously suggested by Horn et al (2011). In addition, to apply foci formation in neutron biodosimetry additional readings at different time-points post-irradiation are desired to detect qualitative differences in foci formations from different ionization densities.…”

“…Chemoradiotherapy (CRT), the concurrent use of radiation therapy and chemotherapy, is a well-established field that in most cases works better than just using either of them alone and, actually contributes to the improvement in the overall health of the patient as well as extending the life expectancy in some patients [11,12]. Decades ago, researchers introduced a theoretical framework to describe the interaction of cytotoxic chemotherapy and radiotherapy [13], called "spatial cooperation", it proposed that the action of radiation and chemotherapeutic drugs is directed toward different target sites in the body and might interaction with each other.…”
