Dose-Dependent Effect of Sphingosine-1-Phosphate in Mouse Oocyte Maturation Medium on Subsequent Embryo Development

Abstract: Aim: To investigate the dose-dependent effect of sphingosine-1-phosphate (S1P) supplementation on meiotic maturation, apoptosis and developmental competence of immature mouse oocytes. Methods: Immature oocytes from 5- to 6-week-old BDF1 mice were incubated in maturation medium containing S1P with six different doses (10 nM, 50 nM, 100 nM, 500 nM, 1 µM, and 2 µM) for 18–19 h. Fertilization and blastocyst formation was assessed. All blastocysts were stained by anti-caspase antibody. Results: Treatment with 100 n… Show more

“…Ceramides, SMs, sphingosine and sphingosine-1-phosphate are involved in reproduction and development in Drosophila [63, 66], C . elegans [65], and mice [67], and based on our data, associated with and used during development of neonatal Daphnia . HR96 regulates several genes in the uptake of lipids and the metabolism of SM ( Fig 7 ), including magro, NPC1b, a cholesterol transporter, and two different sphingomyelinases [44, 60, 68].…”

“…S1P has been implicated in oocyte maturation in mice, where oocytes collected in the presence of S1P showed a consistent and significant delay in oocyte maturation ( Hinckley et al, 2005 ). However, adding S1P to the oocyte maturation medium was found to be beneficial to the development of immature mouse oocytes ( Jee et al, 2011 ). S1P has also been shown to prevent apoptosis of mice oocytes exposed to chemotherapeutic damage ( Morita and Tilly, 2000 ), and it protected bovine oocytes from heat shock during maturation ( Roth and Hansen, 2004 ).…”

“…Immature mouse oocytes were incubated in maturation medium with S1P, the blastocyst formation rate was higher, and the percentage of apoptotic blastomeres was significantly lower, than those of the control group. 66 A recent report indicated that addition of S1P to human oocyte culture medium significantly reduced fertilization rate although such treatment decreased embryo fragmentation. 67 Ceramide decreased the mouse blastocyst formation rate and induced embryonic cell apoptosis, but S1P partly inhibited the effect of Cer during the mouse preimplantation embryonic development.…”
