Dose-Dependent Cytotoxic Effects of Menthol on Human Malignant Melanoma A-375 Cells: Correlation with TRPM8 Transcript Expression

Abstract: Background: Transient receptor potential melastatin 8 (TRPM8), a principle membrane receptor involved in calcium ion influx and cell signal transduction, has been found to be up-regulated in some cancer types, including melanomas. Efficiency of menthol, an agonist of TRPM8, in killing melanoma cancer cells has been reported previously, but the mechanisms remain unclear. We here determined whether in vitro cytotoxic effects of menthol on A-375 human malignant melanoma cells might be related to TRPM8 transcript … Show more

“…Likewise, a 30‐s treatment of stem cells derived from a buccal fat pad and grown on machined titanium disks showed a reduction in cellular viability (Park et al , ). On A‐375 cells, M exerted in vitro cytotoxic effects, with an IC 50 value of 0.012 mM (the M concentration in Listerine® is 2.69 mM) (Kijpornyongpan et al , ). On Hep‐2 cells, the IC 50 value of T was 0.7 mM (the T concentration in Listerine® is 4.3 mM) (Stammati et al , ).…”

“…In this study, three kinds of monoterpenes: thymol, linalool, and menthol, were and menthol exhibited anti-cancer effects against human tumor cell lines, such as prostate cancer [19,20], malignant melanoma [21], colon cancer [22], and gastric cancer [23]. They suppressed cell growth and induced apoptosis in tumor cells in a time-and dose-dependent manner.…”

“…29,135 Chanchao and coworkers had in fact stated an effectiveness comparable with that of 5-FU, with the advantage that menthol has no toxicity, while the side-effects of 5-FU are multiple and non-negligible. 135 Menthol also showed a significant decrease on the viability of a bladder cell line T24, inducing cell death through TRPM8 activation. 136 Inventors from the Miguel Hernández University have patented that the TRPM8 receptor is linked to the control of lacrimation, and that its activation with agonists, like menthol, increased tearing, thus leading to a promising remedial for the dry eye condition.…”
