Genus Diplotaxis is represented by more than 20 species mainly distributed in the North Mediterranean. Out of these Diplotaxis tenuifolia (wild rocket) is used widely in Italian and French cuisine. It has also been used for medicinal purpose for different illnesses throughout the history. In this study the germination behaviour of the seeds of D. tenuifolia was investigated. The seeds were very sensitive to pH and salinity stress. The seed germination increased under daylight conditions reaching a level of 80 percent but decreased in dark and continuous light. A remarkable increase was recorded (60%) in the germination of seeds subjected to +4°C shock for a week or two before left for germination. Pre-hydration followed by re-drying increased the germination to some extent (28%). These findings reveal that the storage conditions are important for establishing a seed bank of D. tenuifolia, in order to get highest germination.

Variations in levels of seed dormancy among conspecific populations are poorly understood. We aim to determine the variations in the level of dormancy in Rosa multibracteata along an elevational gradient, and to analyze the mechanisms underlying seed dormancy. The study was conducted in an arid valley in the eastern Tibetan Plateau at five elevations: 1700, 1900, 2100, 2300 and 2500 m. Achene traits were measured and physiological levels of dormancy were determined by measuring germination percentages of achenes treated with H 2 SO 4 scarification, warm stratification and cold stratification and combinations of those treatments. Achene size, mass and pericarp thickness increased with increasing elevation, but embryo width decreased with increasing in the arid valley. The level of dormancy generally increased with increasing elevation. The level of dormancy of the rose achenes is intermediate from sites at elevations of 1700, 1900 and 2100 m and deep at elevations of 2300 and 2500 m. Comprehensive analysis suggests that the elevational pattern of level of seed dormancy could be ascribed to intrinsically thicker pericarp and deeper physiological dormancy in the embryo, and extrinsically decreased temperature, increased precipitation and soil moisture at the higher elevations. The great variations of achene traits and levels of seed dormancy across elevations suggest a phenotypic differentiation exists within this rose species. Thus, habitat conditions and achene traits should be considered when selecting roses as materials used to propagate seedlings for ornamental purposes or plant restoration.

Dormancy breaking treatments in Northern Wild Rice (Zizania palustris L.) seed suggest a physiological source of Dormancy

Dormancy is a limiting factor for breeding in northern wild rice (NWR; Zizania palustris L). This study developed a dormancy curve and tested a combination of scari cation and hormone treatments, across three timepoints, for their ability to break dormancy in NWR and produce viable seedlings and plants. A dormancy curve was established across 9 months post-harvest, which showed maximum germination (95 %) by 17 weeks post-harvest and high germination (≥81 %) through the rest of the testing period. Next, dormancy breaking treatments were tested. At 1 week post-harvest, few seeds germinated (≤ 15 %) across all treatment combinations. However, sulfuric acid increased germination shortly after harvest (5.8 %), compared to water (0.5 %) and NaClO (0 %) but resulted in stunted seedlings, all but one of which died shortly thereafter. At 7 weeks, sulfuric acid treated seeds did not result in signi cantly higher germination than water and maximum germination was still below 15%. By 11 weeks post-harvest, the water treatments had the highest germination and resulted in the most viable plants, indicating that dormancy had begun to break naturally and exceeded the effect of the other scari cation treatments. Hormonal treatments had no signi cant effect on germination or seed viability and no strong conclusions could be drawn about their effect on seedling or plant health. Due to the inability of early germinated seed to consistently produce viable plants and the increase in germination following su cient cold storage, it is likely that NWR seed has intermediate or deep physiological dormancy.
