Dopaminergic dysfunction in tourette syndrome

Abstract: A prospective clinical and biochemical study on the effects of treatment with haloperidol has been performed in seven patients with Tourette syndrome. Pretreatment cerebrospinal fluid levels of homovanillic acid (CSF HVA) were significantly reduce in all patients, whereas 5-hydroxyindoleacetic acid was reduced in only two. With haloperidol treatment, symptoms decreased in all cases (21 to 88%) and clinical improvement was associated with an increased level of CSF HVA, often returning to the normal range. Optim… Show more

“…Higher scores indicate worsening phenotype. including the measurement of homovanilic acid levels in post-mortem studies (Butler et al, 1979;Singer et al, 1982), it appears that the most direct evidence has been the strikingly consistent increase in AMP-induced DA rel that was first shown via PET imaging in the pilot study mentioned above (Singer et al, 2002). Improved tomographic resolution has allowed us to examine specific subdivisions of the striatum and, in the current study, we have observed significantly elevated AMP-induced DA rel in the ventral striatum.…”

“…Ever since the original DA hypothesis for TS was proposed (Singer et al, 1982), the underlying dopaminergic deficit leading to TS dysfunction has eluded investigators. Thus, despite the many attempts to characterize TS, Motor tics, phonic tics, and impairment were measured using the YGTSS (Leckman et al, 1989).…”

“…The DA hypothesis of TS was originally proposed nearly 30 years ago [617], and has been corroborated by post-mortem data [618]; however the identification of an underlying DA deficit leading to dysfunction in TS has proven to be elusive [619]. Available evidence suggests that a phasic DA imbalance, similar to that seen in schizophrenia, may help to explain the pathophysiology of TS [619,620].…”
