Dopamine transporters govern diurnal variation in extracellular dopamine tone

Abstract: The majority of neurotransmitter systems shows variations in state-dependent cell firing rates that are mechanistically linked to variations in extracellular levels, or tone, of their respective neurotransmitter. Diurnal variation in dopamine tone has also been demonstrated within the striatum, but this neurotransmitter is unique, in that variation in dopamine tone is likely not related to dopamine cell firing; this is largely because of the observation that midbrain dopamine neurons do not display diurnal flu… Show more

“…The microdialysis data in the current study documented decreased tonic extracellular DA levels in SI compared with GH animals at baseline. Extracellular levels of DA in the NAc are determined, in part, by DA transporter (DAT) activity (Ferris et al, 2014). The increased uptake of DA observed in the NAc core and shell of SI rats in the current study and in the NAc core in a previous report suggests that SI leads to augmented DAT levels in this brain region (Yorgason et al, 2015).…”

“…Others showed NA to be of crucial influence to MPTP susceptibility in mice [34,35]. Trends observed in levels of DA, NA and 5-HT between HR and LR might be related to behavioral differences between groups, although diurnal effects could obscure the effects on the DA release of the HR [36]. Individual differences in monoamine levels within species were previously reported [37,38].…”

“…The delayed restoration of release suggests that the amphetamine-induced reexpression of DAT monomers on the membrane may be necessary for, and lead to, increases in the amount of releasable dopamine. For example, it was recently discovered that DAT expression can determine, at least in part, the amount of readily releasable dopamine (Egana et al, 2009;Ferris et al, 2014). However, if DAT expression leads to refilling of vesicles, it is clear that some amount of time longer than 1 h, but less than 24 h, is required for this process.…”
