Dopamine fails to inhibit renal tubular sodium pump in hypertensive rats.

Abstract: We have previously reported that dopamine-1 receptor-mediated activation of phospholipase C is diminished in renal cortical slices of adult spontaneously hypertensive rats. To determine the potential consequences of this phenomenon, we performed the present studies in which renal proximal tubule suspensions obtained from spontaneously hypertensive and Wistar-Kyoto rats of 10-12 weeks of age were used. The tubule suspensions were incubated with dopamine in the presence or absence of dopamine receptor antagonist… Show more

“…14,15 Rats were anesthetized with sodium pentobarbital (50 mg/kg, IP). After a midline incision, selective perfusion of the kidneys was performed with modified Krebs-Hensleit buffer containing collagenase type IV (230 U/mL) and hyaluronidase type III (250 U/mL).…”

“…13,14 The enriched proximal tubules from each group of rats were suspended in modified Krebs-Henseleit buffer A 13 (KHB A). The proximal tubular suspension (1 mg/mL) was incubated with KHB A, dopamine (10 nmol/L), or forskolin (1 mol/L) at 37°C for 15 minutes.…”

“…The tubules were permeabilized by rapid freezing in dry ice-acetone and subsequent thawing. The Na ϩ ,K ϩ -ATPase activity was measured as described earlier 14 and expressed as nanomoles of inorganic phosphate per milligram protein per minute and determined as the difference between total and ouabain-insensitive ATPase.…”
