Donepezil attenuates vascular dementia in rats through increasing BDNF induced by reducing HDAC6 nuclear translocation

Abstract: Vascular dementia (VD) is the second most common dementia disease after Alzheimer’s diseases (AD) in the world. Donepezil is used to treat mild to moderate AD, and it has been shown to treat cognitive impairment and memory deficits caused by VD. However, the action mechanism of donepezil against VD has not been clarified. In this study, a bilateral common carotid artery occlusion (BCCAO) model was established in rats to simulate the pathology of VD. Two weeks after the surgery, the rats were administered donep… Show more

“…Donepezil is a clinically used US FDA‐approved drug for dementia of Alzheimer’s disease. It is also clinically employed for dementia of other etiologies [31,32]. Donepezil is taken as positive standard in our study based on our previous work [33].…”

“…Therefore, the present study has been conducted to investigate the efficacy of ozagrel, a thromboxane A2 synthase inhibitor in rat model of HHcy‐induced VCID. Donepezil, a central cholinesterase inhibitor, currently being clinically used for dementia of Alzheimer’s disease [31] and other etiologies [32] has been employed as a positive standard in the present investigation.…”

“…Donepezil, used to treat mild to moderate AD, has been shown to treat cognitive impairment and memory deficits caused by VD. Jian et al [ 43 ] showed that donepezil treatment significantly improved cognitive performance. Furthermore, donepezil treatment significantly attenuated neurodegeneration and restored synapse dendritic spine density in the cortex and hippocampus.…”
