Domains of Tra1 Important for Activator Recruitment and Transcription Coactivator Functions of SAGA and NuA4 Complexes

Abstract: The Tra1 protein is a direct transcription activator target that is essential for coactivator function of both the SAGA and NuA4 histone acetyltransferase (HAT) complexes. The ϳ400-kDa Saccharomyces cerevisiae Tra1 polypeptide and its human counterpart TRRAP contain 67 or 68 tandem ␣-helical HEAT and TPR protein repeats that extend from the N terminus to the conserved yet catalytically inactive phosphatidylinositol 3-kinase (PI3K) domain. We generated a series of mutations spanning the length of the protein an… Show more

“…Moreover, the interaction of Gcn4 with SAGA in cell extracts was not impaired by the Taf9 CRD mutation. Tra1 and Taf12, two SAGA subunits previously shown to interact with the Gcn4 activation domain (51,63,69), are efficiently coimmunoprecipitated in a complex with Spt7 (Fig. 5D), and the same subunits were also pulled down by Gcn4 (Fig.…”

“…Previous studies have shown that SAGA interaction with the Gcn4 activation domain is mediated by Tra1 (51, 63) and Taf12 subunits (48,63). Moreover, these subunits are also required for transcriptional activation of target genes by Gcn4, thereby providing physical and functional interaction of SAGA subunits with Gcn4.…”

“…As mentioned above, NuA4 is a multisubunit protein complex and is essential for acetylation of histones H4, H2A, and H2A.Z (4-7). NuA4 is targeted to the UAS of the promoter of the target gene by a transcriptional activator (10,11,14,17,46,70). At the ribosomal protein genes, NuA4 is recruited by the activator Rap1p, and such targeted recruitment is essential for transcriptional initiation (10,14,17,70).…”

“…Tra1 is also likely responsible for activator recruitment of the NuA4 coactivator. Most of Tra1 is composed of short repeated motifs, including HEAT and TPR repeats, which are N-terminal to the PI3 kinase-like domain (Knutson and Hahn 2011). Systematic mutation of Tra1 has shown that about two-thirds of the protein is essential for growth.…”
