Does isotretinoin therapy of acne cure or cause depression?

Abstract: Cosmetic defects in acne may provoke a wide range of mental disorders (depressive, social-phobic, etc.). Isotretinoin is a very effective acne treatment; hence, it usually resolves the associated mental disorders. However, more available data show the possible association of taking isotretinoin and the onset of a depressive syndrome that includes frank depression and even suicidal ideation. The frequency of depressive disorders during isotretinoin treatment varies from 1% to 11% in different studies, and it is… Show more

“…The use of isotretinoin has been frequently associated with depression and less frequently with suicide and psychosis [14]. Isotretinoin may disrupt the functions of several cerebral regions like hippocampus, corpus striatum, and frontal cortex, particularly the orbitofrontal cortex, which are associated with the pathophysiology of depression [15].…”

“…As expected, topic INN into the systemic circulation was high, with systemic exposure to INN being approximately 22 times higher than that in the epidermis [ 37 ]. It can across the blood-brain barrier easily and act in the brain tissue [ 38 ]. Since it crosses the blood-brain barrier, INN affects the expression of a wide spectrum of genes in the limbic structures, thus changing the function of the dopaminergic, serotonergic, noradrenergic neurons involved in the modulation of mood [ 38 ].…”

Our results also suggest that INN can induce depressive behavior ( Fig 3 ) , which is associated with the prevetion of hippocampal 5-HT-5-HT1A/1B system and induces apoptosis (Figs 4 and 5 ) .…”

“…Depressive disorders are more of a concern with interferon-α (especially interferon-α2b) than interferon-β (Raison 2005). Isotretinoin is known to be associated with depression, suicidal ideation and psychosis (Borovaya 2013). The frequency of depressive disorders during treatment with this drug varies from 1 to 11% in different studies, but it is unclear whether this is a consequence of isotretinoin therapy (Borovaya 2013).…”
