The activity of the adenylate cyclase located in membranes prepared from hippocampus of adult rat can be stimulated by serotonin (5-HT) (Ka = 4 X 10(-7) M). The maximal effect is obtained with 10 microM 5-HT. Freezing of the tissue decreases the 5-HT stimulation; this stimulation is optimal in the presence of 82.5 mM Tris-maleate buffer (pH 7.4) and 50 microM GTP. The adenylate cyclase activity of membranes prepared from cortex, hypothalamus, and colliculi of adult rats is not significantly stimulated by 5-HT. Dopamine (DA) also stimulates adenylate cyclase located in hippocampal membranes; its effect can be blocked by haloperidol (10(-6) M), which fails to inhibit 5-HT stimulation. Moreover, p-chlorophenylalanine treatment for 2 weeks or selective lesion of 5-HT axons afferent to the hippocampus increases the Vmax of 5-HT stimulation, but fails to change that of DA stimulation. The 5-HT stimulation can be inhibited by metergoline, spiroperidol, and pizotyline (10(-6) M), but not by the same concentrations of mianserin, ketanserine, alprenolol, phenoxybenzamine, and mepyramine. The 5-HT stimulation of adenylate cyclase of hippocampal membranes can be mimicked by tryptamine, 5-methoxytryptamine, bufotenine, and to a lesser extent by LSD; N-methyltryptamine, N-methyltryptophan, and 5-hydroxytryptophan are inactive. Studies with kainic acid suggest that the 5-HT recognition site (5-HT1) linked to adenylate cyclase is located on the membrane of intrinsic hippocampal neurons.

The effects of long-term treatment of rats with alaproclate and amiflamine on the number and kinetics of 5-HT1 and 5-HT2 binding sites were investigated using in vitro receptor binding techniques. Some other studies have reported down-regulatory effects of alaproclate and amiflamine on 5-HT2 binding sites in certain regions of the rat forebrain, but no such effects could be detected in the present study. Induction of a high-affinity binding site for 3H-5-HT after long-term antidepressant treatment, as has been reported elsewhere, was not obtained in the present study. The results are compared to the effects obtained by treatment of rats with para-chloroamphetamine (PCA), which depletes the presynaptic neurons of monoamines. These different types of treatment do not cause any change in the binding properties of the specific 5-HT binding sites. It is thus concluded that such manipulations of the presynaptic 5-HT neurons do not affect the postsynaptic 5-HT1 and 5-HT2 binding sites.

High-affinity [3H]serotonin binding activity has been solubilized from bovine cerebral cortical membranes by using Triton X-100, Tween-80, and octyl-fi-D-glucopyranoside. The present report describes the solubilization of high-affinity [3H]serotonin binding activity from bovine frontal cortical membranes. This procedure, employing a mixture of detergents that can be removed from the solubilized preparation, gives a very high solubilization yield and preserves high specific binding. Preliminary pharmacologic and physical characterizations are presented.MATERIALS AND METHODS Preparation of Bovine Cerebral Cortical Membranes. Bovine cerebral cortex was obtained fresh at a local slaughterhouse. Tissues were dissected and packed in dry ice before transit to the laboratory. All homogenization, solubilization, and purification steps were carried out at 0-4C unless otherwise stated. Frozen bovine frontal cortex was thawed in 0.32 M sucrose buffered with 2.5 mM Tris HCl at pH 7.4 and was homogenized as a 10% (wt/vol) suspension in a Teflon glass grinder. The homogenate was centrifuged at 580 x g for 10 min. The pellet was discarded and the supernatant was centrifuged at 50,000 X g for 20 min. washed in a small volume of 0.32 M sucrose, diluted with 25-28 vol of distilled water, mixed, and recentrifuged at 50,000 X g for 20 min. The pellets were resuspended in a small volume of 0.32 M sucrose solution and incubated for 15 min at 300C. Approximately 10-15 vol of cold distilled water were then added and the suspension was centrifuged at 40,000 X g for 25 min. The opalescent grey-white pellet was resuspended in 3-4 vol of distilled water, sonicated (25 W, 5 sec), diluted with distilled water, and centrifuged at 50,000 X g for 25 min. The pellet was resuspended in distilled water, snap-frozen, and stored at -80°C. This fraction, which consisted primarily of lysed synaptosomal membranes, is designated the "crude" membrane fraction.Protein Determinations.
