DOCK2 sets the threshold for entry into the virtual memory CD8+T cell compartment by negatively regulating tonic TCR triggering

Abstract: The control of cytoskeletal dynamics by Dedicator of cytokinesis 2 (DOCK2), a hematopoietic cell-specific actin effector protein, has been implicated in TCR signaling and T cell migration. Biallelic mutations in Dock2 have been identified in patients with a recessive form of combined immunodeficiency with defects in T, B and NK cell activation. Surprisingly, we show here that certain immune functions of CD8 + T cells are enhanced in the absence of DOCK2. Dock2-deficient mice have a pronounced expansion of thei… Show more

“…In normal homeostasis, the pool of T SCM cells in humans is believed to comprise 2-3% of all circulating T lymphocytes [61,65]. Some CD8+ T MEM cells are virtual memory cells that originate from naïve T cells under strong IL15 signaling but without agonist antigen stimulation [66][67][68]. The role of these cells is debatable, but one theory is that they provide innate-like protection during the earliest stages of bacterial or viral infection [68,69].…”
