DOC2A and DOC2B are sensors for neuronal activity with unique calcium‐dependent and kinetic properties

Abstract: Elevation of the intracellular calcium concentration ([Ca 2+ ] i ) to levels below 1 lM alters synaptic transmission and induces short-term plasticity. To identify calcium sensors involved in this signalling, we investigated soluble C2 domain-containing proteins and found that both DOC2A and DOC2B are modulated by submicromolar calcium levels. Fluorescent-tagged DOC2A and DOC2B translocated to plasma membranes after [Ca 2+ ] i elevation. DOC2B translocation preceded DOC2A translocation in cells co-ex… Show more

“…PC12 cells were incubated with 4 M fura-4 acetoxymethyl ester (AM) (Invitrogen, Eugene, OR) for 45 min in the 37°C incubator before being used for experiments (Groffen et al, 2006). To facilitate loading of the AM ester indicators, the mild detergent pluronic F-127 was added to fura-AM in a 1:1 ratio and vortexed thoroughly with 1 ml of DMEM (Invitrogen) immediately before incubation.…”

“…They performed most of their experiments using a buffer without calcium (i.e., Nonidet P-40 lysis buffer). Since DOC2b has conserved Ca 2ϩ -binding sites and is thought to be a Ca 2ϩ sensor protein in other cell types such as neuron (32) and chromaffin cells (33), Ca 2ϩ might be important for the physiological properties of DOC2b in ␤-cells. Further work is required to uncover the underling mechanisms by which DOC2b regulates SNARE assembly in response to insulin.…”

“…DOC2A is expressed predominantly in the brain and localizes to synaptic vesicles. it has been hypothesized that DOC2A regulates synaptic activity though ca-dependent mechanisms (12). PPP4C encodes a serine/threonine phosphatase that has a role in motor neuron survival (4).…”
