Do the Microbiota Influence Vaccines and Protective Immunity to Pathogens?

Andrew J. Macpherson

Abstract: In contrast to live attenuated vaccines, which are designed to induce immunity through a time-limited bloom in systemic tissues, the microbiota is a persistent feature of body surfaces, especially the intestine. The immune responses to the microbiota are idiosyncratic depending on the niche intimacy of different taxa and generally adapt the host to avoid overgrowth and maintain mutualism rather than to eliminate the organisms of that taxon. Both the microbiota and the host have so much molecular cross talk con… Show more

“…Going forward, rationally designed vaccines leveraging the microbiome as endogenous adjuvant holds promise in the field of vaccinology. Such approaches may require personalization and engineered reversibility to manage efficacy and potential complications in frail individuals [ 116 ]. Given that diet is one of the most potent factors shaping the gut microbiome, nutritional interventions with pre- and probiotics that promote a diverse microbiome to sustain health have received considerable interest as discussed below.…”

“…Yet such applications are not without risk. The endogenous intestinal microbiota can support host immune responses against enteric pathogens but they are also active participants in the induction of immune tolerance and maintenance of mutualism [ 24 ]. Alteration of the microbiota at an early age risks disruption of these roles with the potential induction of (auto)inflammation.…”

“…The immunological basis for how the microbiome may influence vaccination has been a subject of exploration for several years [ 12 , 13 ], and more recent studies have expanded upon this topic [ 10 , 14 , 15 , 16 ]. In this context, a more diverse intestinal microbiota has been shown to be favorable for a good immune response to oral vaccines against intestinal pathogens [ 12 ].…”
