Do not just do it, do it right: urinary metabolomics –establishing clinically relevant baselines

Abstract: Metabolomics is currently being adopted as a tool to understand numerous clinical pathologies. It is essential to choose the best combination of techniques in order to optimize the information gained from the biological sample examined. For example, separation by reverse-phase liquid chromatography may be suitable for biological fluids in which lipids, proteins and small organic compounds coexist in a relatively nonpolar environment, such as serum. However, urine is a highly polar environment and metabolites a… Show more

“…While many analytical tools can be used for the identification and quantification of urine metabolites, mass spectrometry, especially when linked to a suitable separation technique, can serve as an ideal platform for metabolomics research (Dunn et al, 2013;Armitage & Barbas, 2014;Trivedi & Iles, 2014a;Zhang et al, 2012a). MS offers the preferred sensitivity, high throughput capability and the desired quantitative outputs, absolute or relative.…”

“…Numerous methods of urinary metabolomic profiling have been widely reported in order to ascertain changes in human metabolism owing to drug or disease processes (Trivedi and Iles, ). However, during pregnancy there are dramatic metabolic changes which are due to the establishment and growth of the fetus and maternal adaptation to the fetus (Lain and Catalano, ; Abduljalil et al ., ).…”

“…This gestational age and mathematical correction required in our MALDI ToF MS urinary spectra pro iling tests is akin to the multiple of median correction adopted in conventional maternal serum and proposed metabolomics screening tests for Down syndrome [27][28][29][30][31][32]. Thus, an optimized screening https://doi.org/10.29328/journal.apb.1001008 test for Down syndrome pregnancy by maternal urinary MALDI ToF MS analysis will require dating of the pregnancy at sampling so that an appropriate decision algorithm can be applied.…”
