Do gap junctions play a role in nerve transmissions as well as pacing in mouse intestine?

Abstract: Varicosities of nitrergic and other nerves end on deep muscular plexus interstitial cells of Cajal or on CD34-positive, c- kit-negative fibroblast-like cells. Both cell types connect to outer circular muscle by gap junctions, which may transmit nerve messages to muscle. We tested the hypotheses that gap junctions transmit pacing messages from interstitial cells of Cajal of the myenteric plexus. Effects of inhibitors of gap junction conductance were studied on paced contractions and nerve transmissions in small… Show more

“…The question whether or not ICC are involved in neurotransmission has also been investigated by using the hypothesis that neural activation by field stimulation would release neurotransmitters onto ICC and this information would be transmitted to smooth muscle cells via gap junctions. Experimental evidence showed that blockade of gap junctions did not alter neurotransmission (3,14). This is consistent with our view that neurotransmitters are secreted from nerve varicosities directly to smooth muscle cells.…”

“…The disruption of the gap junctions by pharmacological agents has little effect on the slow-wave activity in the circular muscle cells of intact muscle strips (40,43,142). Other studies have demonstrated poor coupling between the gastric ICC-MY and the adjacent circular smooth muscle cells (37,41).…”

“…Further investigations are needed to determine how the slow waves generated independently in the ICC-MY and in circular smooth muscle cells interact to influence the regulation of RPCs in the gut. It is noteworthy that the tight coupling between the ICC and smooth muscle cells, which is an important basis of the pacemaker hypothesis, has been questioned (37,40,43).…”

“…[251][252][253] The gap junction uncouplers, octanol and carbenoxolone, failed to disrupt pacemaker activity in various species, 99,254 however no tests to measure the degree of electrical uncoupling have accompanied these studies. More recently, Daniel et al 255 used peptide analogs of epitopes in the extracellular domain of connexins because these are considered to be specific gap junction uncouplers. Application of these peptide analogs had no effect on paced contractions and nerve transmissions in murine small intestine, but again no tests of electrical coupling were included in the results.…”
