DNMT3B overexpression downregulates genes with CpG islands, common motifs, and transcription factor binding sites that interact with DNMT3B

Abstract: DNA methylation is a key epigenetic modification to regulate gene expression in mammalian cells. Abnormal DNA methylation in gene promoters is common across human cancer types. DNMT3B is the main de novo methyltransferase enhanced in several primary tumors. How de novo methylation is established in genes related to cancer is poorly understood. CpG islands (CGIs), common sequences, and transcription factors (TFs) that interact with DNMT3B have been associated with abnormal de novo methylation. We initially iden… Show more

“…It appears that the methylation level of the IRF1 promoter can still be modified by some additional factors in different cellular contexts. For example, overexpression of the methyltransferase gene DNMT3B led to increased IRF1 gene methylation levels and repression of its expression [68]. In contrast, exposure of cells to the TET1 dioxygenase under IFN-γ-stimulated conditions could demethylate the IRF1 gene and activate its transcription.…”

“…Increased levels of DNMT3B, a de novo DNA methyltransferase, were a recurrent event detected in oral cancer tissues [ 52 , 53 , 54 , 55 ] as well as oral premalignant lesions [ 56 , 57 ], strongly supporting its role in tumorigenesis. In the vicinity of DNMT3B , an amplification hotspot comprising two stemness-related genes, ID1 and BCL2L1 ( Figure 4 C), is commonly found in human embryonic stem cell cultures.…”
