DNase I- and micrococcal nuclease-hypersensitive sites in the human apolipoprotein B gene are tissue specific.

Abstract: We have mapped the DNase I-and micrococcal nuclease-hypersensitive sites present in the 5' end of the human apolipoprotein B (apo-B) gene in nuclei from cells expressing or not expressing the gene. Four DNase I-hypersensitive sites were found in nuclei from liver-derived HepG2 cells and intestine-derived CaCo-2 cells, which express the apo-B gene, but not in HeLa cells, which do not. These sites are located near positions -120, -440, -700, and +760 Apolipoprotein B (apo-B) is the major protein component of … Show more

“…Some of the identified AT islands have been experimentally shown to function as MARs. Examples include the c-Myc MAR (29) and the ApoB MAR (30). Our preliminary results confirm also that the Z79699/AF385609 AT island, the "hottest" identified region for bizelesin binding motifs, binds specifically to the isolated nuclear matrix, and hence functions as an in vitro MAR.…”

“…However, the AT islands in GenBank™ sequences X04682 and X00364 correspond precisely to the known AT-rich MAR domains in the apoB and c-myc genes, respectively (29,30). The potential bizelesin targets also include AT islands in the AT-rich fragile sites (31), FRA16D (in Gen- Bank™ AF217490, with a total of 60 bizelesin cross-linking sites, Table II) and FRA16B (see on-line supplement).…”

“…Tissue Culture and Transient Transfection Assays-Human hepatoma cells (HepG2), Chinese hamster ovary (CHO) cells, and HeLa cells were grown as described previously (18). Transient transfections with the various plasmid constructs were performed by the calcium phosphate coprecipitation method as described previously (19) with 7 g of the CAT gene expression plasmid and 6 g of an internal reference plasmid (pRSV␤-gal).…”
